摘 要:马铃薯(Solanum tuberosum L.)是一种对盐中度敏感的作物,由于土壤盐渍化问题,严重限制了马铃薯产业的发展。本试验研究离体条件下盐胁迫对二倍体马铃薯富利亚(S.phureja,PHU)和窄刀薯(S.stenotomum,STN)杂种(PHU-STN)无性系相关生理指标的影响,以期探讨盐胁迫下二倍体马铃薯耐盐的生理机制。从45份PHU-STN中,根据其在盐胁迫下6项生长参数(芽长、根长、芽鲜重、根鲜重、芽干重、根干重)的表现选出5份耐盐无性系、5份中耐盐无性系、5份感盐无性系建立耐盐程度不同的三个组,以四倍体品种Bintje为耐盐对照,Mainechip为感盐对照,用浓度为30mmol/L的NaCl溶液进行不同天数的胁迫,测定叶片质膜透性、丙二醛和脯氨酸含量。结果表明,随着盐胁迫天数的延长,试管苗的叶片质膜透性及丙二醛含量的相对值逐渐升高,二倍体组间差异极显著;随着盐胁迫天数的延长,脯氨酸含量相对值的变化呈先升高后降低的趋势,二倍体组间差异极显著。因此,叶片质膜透性、丙二醛含量、脯氨酸含量可以作为二倍体马铃薯耐盐性鉴定的生理指标。As a moderately salt sensitive crop, the development of potato (Solanum tuberosum L. ) industry is sen- ously hindered due to soil salinization. In this study, S. phureja (PHU) and S. stenotomum (STN) hybrid ( PHU- STN) clones were evaluated in vitro for their physiological indices under salt stress in order to investigate mecha- nisms of stress tolerance in diploid potatoes. Three groups ( five salt-tolerant, five moderate salt-tolerant, and five saltsensitive clones)selected out based on their performance fresh weight, root fresh weight, shoot dry weight, and root of six growth parameters (shoot length, root length, shoot dry weight) from 45 PHU-STN clones were evaluated for their membrane permeability, malondialdehyde, and proline content at various days after 30mmol/L NaC1 stress with tetraploid variety Bintje as salt-tolerant control and Mainechip as salt-sensitive control. With days after salt stress go- ing,relative leaf membrane permeability, and malondialdehyde content increased, and differences were significant (P 〈0.01 ) among the three groups. Relative proline content was increased and then decreased with days after salt stress going,and differences were also significant( P 〈 0. 01 )among the three groups. These results indicated that leaf membrane permeability, malondialdehyde content, and proline content could be used as physiological indices for evaluating salt tolerance of diploid potatoes.
