摘 要:目的观察子宫颈鳞状细胞癌组织中β-连环蛋白(catenin)和细胞周期蛋白D1(Cyclin D1)的表达变化,并探讨其临床意义。方法采用免疫组化法检测58份子宫颈鳞状细胞癌组织及22例正常子宫颈组织中β-catenin和Cyclin D1的表达,分析两者之间及其与子宫颈鳞状细胞癌临床病理参数的关系。结果子宫颈鳞状细胞癌组织中β-catenin异位表达率及Cyclin D1阳性表达率均显著高于正常子宫颈组织(P均<0.05);子宫颈鳞状细胞癌组织中β-catenin和Cyclin D1的表达呈正相关,P<0.01(r s=0.538);β-catenin表达与子宫颈鳞状细胞癌病理分级及淋巴结转移有关(P均<0.05),Cyclin D1表达与子宫颈鳞状细胞癌病理分级、临床分期及淋巴结转移有关(P均<0.05)。结论β-catenin异常表达所致Cyclin D1激活表达上调可能是子宫颈鳞状细胞癌发生、发展的重要途径,联合检测两者表达有助于评估肿瘤恶性程度、侵袭转移能力及为预后分析和临床治疗提供依据。Objective To detect the expression of β-catenin and Cyclin D1 in cervical squamous cell carcinoma tissue and to investigate its clinical significance. Methods The expression of β-catenin and Cyclin D1 was detected by immuno- histochemistry method in 58 cases of cervical squamous cell carcinoma tissues and 22 cases of normal cervical tissues, then the relationship between the expression of β-catenin and Cydin D1 and the clinicopathological parameters of cervical squa- mous cell carcinomas was also analyzed. Results The rates of β-catenin ectopic expression and Cyclin D1 positive expres- sion were both obviously higher in cervical squamous cell carcinoma tissues than those in normal cervical tissues ( all P 〈 0.05) ; the expression of β-catenin was positively correlated with that of Cyclin D1 in cervical squamous cell carcinomas (r1 = 0.538, P 〈 0.01 ) ; the expression of β-catenin was correlated with the pathologic grades and lymph node metastasis ( all P 〈 0.05 ) ; and the expression of Cyclin D1 expression was correlated with the pathologic grades, clinical stages and lymph node metastasis ( all P 〈 0.05). Conclusions Cyclin D1 activate the up-regulated expression due to β-catenin ab- normal expression, which may play an important role in the occurrence and development of cervical squamous cell carcino- ma; the combined detection of β-catenin and Cyclin D1 can be helpful to assess the malignancy degree, the characteristics of invasion and metastasis; and it can be used to provide the basis for prognostic analysis and clinical treatment.
