摘 要:在有机相中以Candida rugosa脂肪酶催化正丁酸和醇之间的酯化反应,用已知水活度的盐/盐水合物对控制反应体系的水活度.在其他条件恒定时,筛选出最适底物醇为正丁醇,最适反应溶剂为正庚烷;考察了盐对的加入量以及盐对的比例对反应过程的影响,并进一步研究了水活度对酯化反应的影响.结果表明,用盐/盐水合物对控制反应体系水活度是可行的,反应的初速度和转化率随水活度的增加而增加,在水活度为0.76时,转化率达到72.43%.In this study, the esterification of n-butyric acid in organic solvent catalyzed by Candida rugosa lipase was investigated when the water activity of reaction system was controlled using different salt/salt hydrate pairs. The optimal substrate alcohol and solvent were screened for the reaction and determined to be n-butanol and n-heptane. The influence of the amount and ratio of salt hydrate pairs on reaction course was studied, and then the conversion of n-butyric acid was detected under different water activities. It is verified that water activity is a good parameter to describe the effect of water on enzyme activity in nonaquaous system. The conversion rate and the initial rate of reaction increase with the increase of water activity. When the water activity was 0.76, the maximum conversion rate was 72.43%.
