摘 要:目的:从四物汤及其组方药味、药对、药组中主要成分的定量比较分析四物汤组方配伍的物质组成特点,为四物汤组方配伍规律的研究提供依据。方法:采用HPLC/DAD法测定,色谱柱:Waters SunFire-C18柱(4.6mm×250mm,5μm);柱温:(30±1)℃;流动相:黄乙腈(A)-0.1%磷酸水(B);流速:0.8mL/min;梯度洗脱。结果:药对当归-川芎,当归-白芍,当归-熟地黄和白芍-川芎中阿魏酸和洋川芎内酯I的含量均高于其组成单味药;药对当归-白芍,白芍-川芎,白芍-熟地黄中芍药苷含量均高于单味药白芍;四物汤全方中阿魏酸,洋川芎内酯I和芍药苷的含量均高于组方单味药;没食子酸和绿原酸在组方前后的含量则没有特定规律;药味,药对,药组和全方中5-羟甲基糠醛(5-HMF)含量均低于单味药熟地黄。结论:四物汤及其药对的配伍具有促进或抑制所含成分溶出的作用,为进一步揭示四物汤功效物质基础及组方配伍特点提供科学依据。Quantitation some major components in Siwu Decoction and its composite medicine, medicine- pairs and medicine-groups to provide evidence for the illustration of Siwu Decoction's compatible regularity. Methods: HPLC/ PDA was used. The column was Waters sunfire-C18 (4.6mm×250mm, 5μm) and the column temperature was set at (30±1)℃; The mobile phase was methyl cyanides (A)-0.1% phosphoric acid aqueous solution (B) and the flow rate was set at 0.8mL/min with gradient elution. Results: The contents of ferulic acid and senkyunolide I in medicine-pairs of Angelicae Sinensis Radix (ASR)- Chuanxiong Rhizoma (CR), ASR-Paeoniae Radix Alba (PRA), ASR-Rehmanniae Radix Praeparata (RRP), PRA-CR all increased compared with the single medicine of ASR, CR, PRA and RRP; The contents of ferulic acid, senkyunolide I and peoniflorin in Siwu decoction were higher than that of single medicine; The contents of gallic acid and chlorogenic acid in single medicine, medicine-pairs, medicine-groups and Siwu Decoction exhibited undefined regularity; The single medicine of RRP exhibited the most content of 5-HMF than medicine-pairs, medicine-groups and the whole formula. Conclusion: The compatibility of the formula and medicine-pairs had the effect of promoting or inhibiting the dissolution of some major components. The present research provided a reliable evidence for the illustration of chemical basis and compatibility regularity of Siwu Decoction.
