控释GDNF对BrdU标记的骨髓MSCs源性前体神经细胞在体内分化的影响

摘  要:目的评价BrdU标记骨髓间充质干细胞(Bone marrow mesenchymal stem cells,BMSCs)用于体内示踪的效果,及控释胶质细胞源性神经营养因子(glial cell line-derived neurotrophic factor,GDNF)干预对移植细胞分化的影响。方法应用免疫双重标记技术对前期关于治疗猴脊髓损伤实验研究的相关的石蜡标本进行回顾性研究。控释GDNF+细胞移植组4例,单纯细胞移植组织4例,分别进行BrdU-Nestin双重染色、BrdU-NF双重染色、BrdU-GFAP双重染色。结果猴脊髓组织石蜡标本内可以显示BrdU阳性细胞,并且可以同时呈NF或GFAP阳性表达,Nestin呈阴性表达;添加控释GDNF者BrdU阳性细胞数量多(P=0.10),并且NF阳性表达水平高(P<0.05),自体神经元保存数量较多(P<0.05)。结论 BrdU体外标记干细胞进行体内示踪研究是一种有效的方法;控释GD-NF可以增强移植细胞的存活、增强其向神经元分化的能力,并且可以更加有效的保护自体神经元,促进神经功的恢复。Objective To investigate the value of BMSCs tracer method by BrdU in vivo,and the effects of controlled release GDNF to the differentiation of transplanted cells.Methods Two groups of paraffin specimens about spinal cord injury experimental study of macaca mulatta were collected,one group treated by neural precursor cells derived from BMSCs and controlled release GDNF,another group treated only by cells,each group including 4 cases.Immune double staining was applied,including BrdU and Nestin,BrdU and NF,BrdU and GFAP,to show transplanted cells trace and differentiation.Results BrdU and NF or GFAP double positive cells could be observed in both paraffin specimens groups,whereas,Nestin was negative.More BrdU positive cells(P = 0.10) and much stronger NF expressing level(P 0.05) could be found in combined transplantation group.Besides,more survival neurons also could be found(P 0.05).Conclusion BrdU marking method for tracing stem cells in vivo is a kind of effective method.Controlled release of GDNF can enhance the survival of the transplanted cells and native neurons,and enhance transplanted cells differentiation potential to neurons,so as to be more effective to promote nerve function rehabilitation.
