摘  要:目的初步探讨白花丹参根水提物作为药理性预处理物的可行性及其神经保护作用机制。方法将健康成年Wistar大鼠随机分为白花丹参高、中、低浓度组和空白对照组、假手术组,分别经白花丹参高、中、低浓度水提物和蒸馏水连续灌胃7d,制备大脑中动脉闭塞模型后24h,采用免疫荧光显微镜检测大鼠脑组织标本凋亡相关蛋白Bax及Bcl-2的表达。结果神经功能评分显示空白对照组评分高于各浓度给药组,低、中、高浓度组与空白对照组比较有统计学意义(P<0.05)。各浓度组间差异无统计学意义(P>0.05)。Bcl-2蛋白阳性染色免疫荧光检测显示:假手术组荧光强度极低,空白对照组较假手术组表达明显增多,有统计学意义(P<0.01);空白对照组荧光强度较低,白花丹参各浓度组与空白对照组相比,荧光强度增高,有统计学意义(P<0.05)。随白花丹参浓度增高,荧光强度逐渐增高,各浓度组间有统计学意义(P<0.05)。Bax蛋白阳性染色免疫荧光检测显示:假手术组荧光强度极低,空白对照组较假手术组表达明显增多,有统计学意义(P<0.01);空白对照组荧光强度强,与空白对照组比较白花丹参各浓度组荧光强度降低,有统计学意义(P<0.05);白花丹参各浓度组随浓度增高,荧光强度逐渐降低,各浓度组间有统计学意义(P<0.05)。结论不同浓度白花丹参水提物预处理均可以明显减少凋亡细胞数,对局灶性脑缺血神经细胞具有保护作用。作用机制可能与增加Bcl-2表达,降低Bax表达有关。Objective To explore protective effects of salvia miltiorrhiza bge.f.alba pharmacological preconditioning on the neurons and its mechanism.Methods Rats were randomly divided into five groups: including salvia miltiorrhiza bge.f.alba(0.5,1.0 and 2.0g/kg),sham-operated and model groups.The drug was administered through intra-gastric method once a day for seven consecutive days.Cerebral ischemia model was induced by occluding middle cerebral artery(MCAO).Neurological evaluation was performed 24h after the establishment of MCAO model and immunofluorescence staining were employed to check the protein expression of BCL-2,Bax for marking the apoptosis of cortical neurons.Results Neurological responses of salvia miltiorrhiza bge.f.alba groups improved significantly compared with the model group.The expression of Bcl-2 and Bax were decreased in sham-operated group.Compared with the model group,the fluorescence strength of the Bcl-2 expression was high in salvia miltiorrhiza bge.f.alba group,while Bax expression was weak(P 0.05).Conclusion Preconditioning by different aqueous extract concentrations of salvia miltiorrhiza bge.f.alba improved neurological performance and modulated neuronal apoptotic.The mechanism may be related with increased Bcl-2 expression and decreased Bax expression.
