金黄色葡萄球菌影响小鼠主要组织相容性复合物I类分子的实验研究

摘  要:目的:研究金黄色葡萄球菌对小鼠主要组织相容性复合物I类分子(MHCI)基因表达的影响。方法:30只BALB/c小鼠随机分成两组,对照组10只,实验组20只,实验组20只小鼠于实验前1天皮下注射金黄色葡萄球菌,实验持续8天。实验结束后取肝脏、脾脏、肾脏和肺组织少许提取RNA,RT-PCR测定MHCI类基因mRNA的表达,Westem印迹测定它们的蛋白表达。结果:第8天实验结束时,肝脏、脾脏、肾脏和肺组织的MHCI类基因mR.NA表达分别为4.35、3.86、4.61和4.23,明显高于对照组;MHCI类基因蛋白表达分别为7.12、7.23、8.65和9.83.明显高于对照组。结论:金黄色葡萄球菌能明显升高机体MHcI基因和蛋白的表达。Objective : To study mouse MHC I expression affected by Staphylococcus aureus. Methods : Thirty BALB/c mouse were randomly divided into two groups, ten served as the control group, twenty in experimental group. The experiment group subcutaneous injected Staphylococcus aureus one day before experiment. The experiment lasted eight days. When the experiment were end, the mice were killed and their livers, spleens, kidneys and lungs were taken out. total R.NA was extracted. I:kT-PCR. and Western blotting were used to examine the mRNA and protein expression of MHC class I gene.aesu[ts : When the experiment was end, liver, spleen, kidney and lung MHC class I gene mR.NA expression level in experimental group were4.35, 3.86, 4.61 and 4.23 respectively, showed obviously higher than in control group, MHC class I gene protein expression level in experimental group were 7.12, 7.23, 8.65 and 9.83 respectively, showed obviously higher than in control group.Conclusions : Staphylococcus aureus can significantly increase organism MHC I gene and protein expression.
