摘  要:目的研究色素上皮衍生因子(pigment epithelium derived factor,PEDF)腺病毒载体注射后,对糖尿病视网膜病变大鼠视网膜新生血管的抑制作用。方法构建、扩增及纯化PEDF病毒载体,注射入2型糖尿病模型大鼠玻璃体中,运用免疫组织化学、PAS染色及HE染色检测PEDF的表达,与对照组比较观察PEDF的表达水平,以及其对视网膜新生血管的抑制作用。结果注射PEDF病毒载体后,PEDF表达量与对照组比较在各个时期均明显增多,同时PAS染色及HE染色均可发现注射组其视网膜新生血管较对照组明显减少。结论注射PEDF腺病毒载体后,稳定表达的PEDF具有抑制新生血管生成,促进异常新生血管消退的作用。Objective To study the inhibition of pigment epithelium derived fac- tor(PEDF) on neovascularization in rats with diabetic retinopathy after the injection of PEDF adenovirus carrier(Ad-PEDF). Methods PEDF viral vectors were structured, amplificated and depurated in vitro and then they were injected into rats' vitreum with di-diabetes mellitus. PEDF expression was detected, the expression level and their inhi- bition on neovascularization were obeserved by comparing with the control group via immunohistochemistry,PAS staining and HE staining. Results In the group treated with Ad-PEDF, compared with control group, the PEDF expression markedly increased at all time points, and retinal neovascularization significantly reduced compared with control group via PAS staining and HE staining. Conclusion After the injection of Ad-PEDE, PEDF can suppress the formation and promote the regression of retinal neo- vascularization in rats with diabetic retinopathy.
