摘 要:目的探讨氟他胺对乳腺癌HCC1937细胞增殖和凋亡的影响及可能的作用机制。方法采用四甲基偶氮唑蓝(MTT)法检测不同浓度(5、10、20μmol/L)的氟他胺对人乳腺癌HCC1937细胞体外生长的抑制作用;Annexin V-FITC/PI流式细胞仪检测细胞凋亡情况;蛋白印迹法(Western blotting)检测核因子κB(NF-κB)和半胱天冬酶(Caspase-3)的蛋白表达水平。结果不同浓度氟他胺作用于HCC1937细胞12、24、36、48h后,细胞生长受到抑制且呈时间和剂量依赖性。不同浓度氟他胺作用HCC1937细胞24、48h后,可显著促进细胞凋亡,并呈时间和剂量依赖性。氟他胺作用HCC1937细胞24h后,随着药物浓度的增加,NF-κB蛋白表达水平逐渐下降,而Caspase-3蛋白表达水平逐渐升高。结论氟他胺能够抑制HCC1937细胞增殖和促进其凋亡,其作用机制可能是通过上调Caspase-3和下调NF-κB蛋白表达实现的。Objective To investigate the potential anti-proliferative and pro-apoptotic effects of flutamide on human breast cancer cell line HCC1937 in vitro and explore its possible anti-cancer mechanism. Methods Different concentration of flutamide (5, 10,20μmol/L) were added to HCC1937 cells. The MTT assay was performed to reveal the inhibitory effect on cell proliferation. The cell apoptosis was determined by flow eytometry using annexin V-FITC/PI staining and the expressions of nuclear faetor(NF-KB) and caspase-3 were examined by Western blotting analysis. Results Flutamide with different concentration treatment inhibited the prolifer- ation and induced the apoptosis of human breast cancer cell line HCC1937 in a time- and dose-dependent manner. The expression of NF-KB protein decreased and caspase-3 protein increased after flutamide treatment for 24h. Conclusion Flutamide can inhibit the proliferation of human breast cancer cell HCC1937 and induce its apoptosis, with the possible mechanisms of dysregulation of NF-KB and caspase-3 expression.
