摘  要:目的:探讨曲古霉素A(Trichostatin A,TSA)处理对胶质瘤细胞系U251增殖和侵袭的调节作用。方法:分别将浓度为200ng/ml、400ng/ml和800ng/ml的TSA加入胶质瘤细胞系U251中。Western blot法检测乙酰化组蛋白H3和H4表达。MTT法检测不同浓度处理组中细胞的增殖能力。Transwell细胞侵袭实验检测不同浓度处理组和对照组中细胞侵袭能力。结果:TSA处理后的U251细胞中乙酰化组蛋白H3和H4表达明显上调(P<0.01)。MTT结果显示U251细胞经TSA处理后的增殖能力明显减弱(P<0.01),且具有剂量依赖性。Transwell细胞侵袭实验显示TSA处理后,U251细胞的侵袭细胞数明显减少(P<0.01),也具有剂量依赖性。结论:TSA可以比较有效地抑制胶质瘤细胞系U251的增殖和侵袭能力,为临床治疗胶质瘤提供了新思路。Objective:The chemical therapy is a hot spot of glioma study.It has been reported that abnormal histone modifications in glioma(deacetylation) correlates with the carcinogenesis and progression of glioma significantly.The purpose of this study is to explore the effect of Trichostatin A(TSA) treatment on the proliferation and invasiveness of glioma cell line U251.Methods:Different concentration of TSA(200ng/ml,400ng/ml and 800ng/ml) was added into glioma cell line U251 and acetylated histone H3 and H4 expression was detected by Western blot.Cell proliferation and invasion was detected by MTT assay and Transwell cell invasive assay.Results:Expression of acetylated histone H3 and H4 were increased significantly in U251 cells after TSA treatment(P 0.01).MTT assay showed that the proliferative capacity of U2 5 1 cells was down-regulated significantly with a dose-dependent manner(P 0.01).Transwell cell invasive assay presented that the number of invasive cells of U251 cells was decreased significantly with a dose-dependent manner(P 0.01).Conclusion:TSA treament could inhibit he proliferation and invasiveness of glioma cell line U251 effectively.The result provides a new idea for the clinical treatment of glioma.
