摘 要:目的:探讨石见穿氯仿及乙酸乙酯部位在体外对血管内皮细胞生物学行为的影响。方法:培养人脐静脉内皮细胞(HUVEC)至3-6代,采用MTT实验、Transwell小室迁移实验、体外小管形成实验观察石见穿氯仿及乙酸乙酯部位对HUVECs体外增殖、迁移、小管形成能力的影响。结果:3.125-25μg/ml的石见穿氯仿部位及乙酸乙酯部位作用于HUVEC 48h,增殖抑制率分别为17.36%-53.08%、9.53%-31.36%,而作用于人肺癌细胞A549增殖抑制率分别为7.10%-31.18%、2.24%-21.69%,两者相比有明显差异细胞毒作用(P=0.001)。3.125-25μg/ml的石见穿氯仿及乙酸乙酯部位作用HUVECs 12h后,迁移抑制率分别为5.39%-57.38%、1.92%-37.37%。3.75-25μg/ml石见穿氯仿部位作用24h时HUVEC小管形成数目减少且管腔不完整,相同浓度石见穿乙酸乙酯部位抑制作用较弱。结论:石见穿氯仿部位抑制内皮细胞增殖、迁移和小管形成作用明显优于乙酸乙酯部位,为石见穿发挥抗血管生成的主要作用部位。Objective:To investigate the effects of the effective parts of Salvia chinensis of the biological behaviors of vascular endothelial cells.Methods:Human umbilical vein endothelial cells(HUVECs) were cultured primarily in vitro.The MTT assay was used to observe the effect of the effective parts of Salvia chinensis on HUVECs and human lung carcinoma A549 cell.Transwell cabin test and tube formation assay were used to observe the effect of the effective parts of Salvia chinensis on the migration and tube formation ability of HUVECs.Results:3.125-25μg/ml chloroform and ethyl acetate extract from Salvia chinensis treated in HUVEC 48h,the proliferation inhibition rate was 17.36% to 53.08%,from 9.53% to 31.36%.Acting on sensitive cell lines of human lung cancer A5 4 9 proliferation inhibition rate of 7.10% to 31.18%,2.24% to 21.69%,both were significantly different in cytotoxicity(P = 0.001).3.125-25μg/ml chloroform and ethyl acetate extract from Salvia chinensis treated the HUVECs 12h migration inhibition rates were 5.39% and 57.38%,1.92% to 37.37%;3.75μg/ml to 25μg/ml trichloromethane extraction part from Salvia chinensis for 24h HUVEC tube formation number reduced,and the lumen was not complete,the same concentration Iwami wear ethyl acetate extract weak inhibition.Conclusion:The trichloromethane extraction part from Salvia chinensis inhibit endothelial cell proliferation,migration and tube formation was significantly better than the ethyl acetate extract for the Salvia chinensis to play a major site of action of anti-angiogenic.
