摘  要:目的探讨研究凋亡抑制基因Livin-α与膀胱癌的易感性关系。方法采用逆转录PCR方法扩增Livin-α基因片段,并对扩增产物进行测序分析,经过比对之后统计分析膀胱癌患者中Livin-α等位基因的数量和出现的频率。根据Livin-α不同的等位基因与患者肿瘤临床分期、病理分级等进行统计学分析。结果 80例膀胱癌患者中Livin-α基因的阳性率为55.0%,对照组组织中Livin-α基因的阳性率为1.5%,二者差异有统计学意义。从所有患者的Livin-α基因测序结果中共发现3种等位基因,统计分析结果表明Livin-αⅠ、Ⅱ和Ⅲ等位基因与患者的肿瘤临床分期和病理分级有统计学意义,而与年龄、性别等因素无统计学意义。结论 Livin-α基因在膀胱癌的发生发展中具有重要意义,并且Livin-α等位基因Ⅰ和Ⅱ在较严重的膀胱癌患者中具有意义。Objective To investigate and study the relationship between apoptosis suppressor Livin-α and susceptibility of bladder cancer.Methods The fragments of Livin-α gene were amplified by reverse transcription PCR,and the sequencing analysis of amplification product was conducted.The numbers and frequencies of Livin-α gene in patients with bladder cancer were statistically analyzed after comparing the results of sequencing analysis.The statistical anal ysis was conducted according to the different alleles of Livin-α,clinical stages of tumor,and pathological classification of tumor.Results The positive rate of Livin-α gene in 80 patients with bladder cancer was 55.0%,while 1.5% in control group.The statistically significant difference was showed.Three kinds of alleles were found in sequencing results of Livin-α gene of all patients.The statistical analysis results indicated that the Livin-α alleles Ⅰ,Ⅱ and Ⅲ had statistical significance with clinical stages and pathological classification of patients with bladder cancer,but no statis tical significance with factors such as age and gender.Conclusion Livin-α gene has important significance in the occurrence and development of bladder cancer,and Livin-α alleles Ⅰ and Ⅱ has significance in serious patients with bladder cancer.
