环氧化酶-2和基质金属蛋白酶-9在骨肉瘤组织中的表达及其临床意义

摘　　要：目的观察环氧化酶-2(Cox-2)和基质金属蛋白酶-9(MMP-9)在骨肉瘤组织中的表达及其临床意义。方法应用免疫组织化学(SP)法,检测35例骨肉瘤、14例骨软骨瘤中Cox-2和MMP-9蛋白的表达情况,并分析其在骨肉瘤不同病理分级、临床分期中的表达及两者之间的相互关系。结果 Cox-2蛋白染色主要表达于细胞浆内,在骨肉瘤中阳性表达率高于在骨软骨瘤中阳性表达率(P<0.05)。MMP-9蛋白染色主要表达于细胞膜和(或)细胞浆内,在骨肉瘤中阳性表达率高于在骨软骨瘤中阳性表达率(P<0.05)。Cox-2和MMP-9蛋白在骨肉瘤组织中的表达与Enneking分期密切相关。Enneking分期Ⅲ期骨肉瘤的Cox-2和MMP-9蛋白的表达强度与Ⅱa期、Ⅱb期比较差异有统计学意义(P<0.05),而Ⅱa期与Ⅱb期的表达强度比较差异无统计学意义(P>0.05)。35例骨肉瘤标本中有23例(65.7%)Cox-2和MMP-9蛋白的表达同时呈阳性,4例(11.4%)同时呈阴性;8例(22.8%)Cox-2和MMP-9蛋白的表达呈不一致性。Cox-2和MMP-9蛋白在骨肉瘤组织中的表达呈正相关。结论 Cox-2和MMP-9蛋白在骨肉瘤组织中均为高表达,提示它们可能对骨肉瘤的发生、发展起重要作用。Objective To investigate the role of Cox-2 and MMP-9 in osteosarcoma by analyze their expression.Methods The expression of Cox-2 and MMP-9 specimens of 35 osteosarcoma and 14 osteochondroma tissues was evaluated by immunohistochemistry.Results Cox-2 protein staining was mainly expressed in the cytoplasm,the positive expression in osteosarcoma was higher than positive expression rate in osteochondroma（P 〈0.05）.MMP-9 protein staining was mainly expressed in the cytomembrane and cytoplasm,the positive expression in osteosarcoma was higher than positive expression rate in osteochondroma（P 〈0.05）.The expression of Cox-2 and MMP-9 in osteosarcoma was correlated with Enneking Staging.There was statistical significance between Enneking stage Ⅲ,Ⅱa and Ⅱb on expression of Cox-2 and MMP 9 protein（P 〈0.05）.There was no statistical significance between Enneking stage Ⅱa and Ⅱb on expression of Cox-2 and MMP-9 protein（P 〈0.05）.There were 23 positive expression of Cox-2 and MMP-9 in 35 osteosarcoma cases,4 negative expression of Cox-2 and MMP-9,8 cases showed expression of inconsistency in Cox-2 and MMP-9.The expressions of Cox-2 and MMP-9 in osteosarcoma were positively correlated（P 〈0.05）.Conclusions Cox-2 and MMP-9 were highly expressed in osteosarcoma.It seems that Cox-2 and MMP-9 contribute to development of osteosarcoma.
