摘  要:目的观察汉黄芩素对小鼠腹水型肝癌的抑制作用,并探讨其作用机制。方法以不同浓度汉黄芩素处理小鼠腹水型肝癌H22细胞,采用MTT法检测汉黄芩素对细胞增殖的抑制作用,采用流式细胞术检测汉黄芩素对细胞周期及细胞凋亡的影响;建立小鼠H22移植肝癌模型,分别给予汉黄芩素(汉黄芩素组)和生理盐水(对照组)腹腔注射10 d,观察实体瘤生长情况。结果汉黄芩素抑制小鼠腹水型肝癌H22细胞增殖、促进细胞凋亡,而且作用强弱呈一定的剂量依赖性;汉黄芩素组处于S期细胞比例上升,处于G0/G1期和G2/M期细胞比例下降;与对照组比较,汉黄芩素组小鼠H22实体瘤质量明显下降(P<0.05或<0.01),且抑瘤率与药物剂量呈正比。结论汉黄芩素不仅对体外培养肝癌H22细胞有直接的细胞毒作用,而且对体内肝肿瘤生长也有抑制作用;上述作用可能与其诱导肿瘤细胞凋亡、干扰细胞周期有关。Objective To observe the inhibitory effect of wogonin on the mouse ascites hepatocarcinoma and to discuss its mechanism of action.Methods The mouse ascitic hepatoma H22 cells were treated by different concentrations of wogonin;the inhibitory effect of wogonin on cell proliferation was detected by MTT method,and the influence of wogonin on cell cycle and apoptosis was detected by flow cytometry.The model of H22 transplanted hepatocellular carcinoma in mice was established and treated with wogonin(wogonin group) and normal saline(control group) intraperitoneally for 10 d the growth of solid tumor in each group was observed.Results Wogonin inhibited the proliferation of the mouse ascitic hepatoma H22 cells,promoted apoptosis,and the strength of its function was certainly dose-dependent;the proportion of cells was increased in the wogonin group in S phase and the proportion of cells was decreased in the G0/ G1 phase and G2/ M phase.Compared with the control group,the quality of solid tumors of the mouse H22 in the wogonin group was decreased significantly(P 0.05 or 0.01),and the anti-tumor rate was proportional to drug dose.Conclusion Wogonin not only has direct cytotoxic effect on H22 tumor cells but also inhibits tumor growth.This might be related to the induction of tumor apoptosis and disturbing the cell cycle.
