摘  要:目的检测卵巢癌细胞株及紫杉醇化疗后卵巢癌组织中WNT-1及FZD-1基因蛋白表达的变化及其意义。方法选用卵巢癌紫杉醇耐药细胞株SKOV3/TAX及敏感细胞株SKOV3,及58例卵巢癌组织,包括术后紫杉醇化疗后复发患者19例(化疗组),术前未化疗患者39例(未化疗组)。采用免疫组化二步法,检测卵巢癌组织及细胞中WNT-1及FZD-1基因的蛋白表达。结果 1.卵巢癌组织中:化疗组WNT-1及FZD-1的蛋白表达光密度值较未化疗组明显降低(分别为30931.34±16420.04 vs 45917.65±12015.39;38625.20±19689.97 vs 55585.82±26330.13),以上差异均有统计学意义(P<0.05);光镜下观察耐药细胞株中WNT-1及FZD-1蛋白表达强度均明显低于敏感株;2.化疗组WNT-1蛋白表达在子宫内膜样腺癌中明显低于浆乳癌(19215.88±6506.49 vs 43193.53±16803.05,P=0.014);未化疗组各基因蛋白表达在年龄、临床分期,分化程度和病理类型中均无显著性差异。3.总体上FZD-1与WNT-1蛋白表达呈显著正相关(r=0.479,P=0.000),化疗组及未化疗组中WNT-1与FZD-1蛋白表达也均呈正相关(分别为r=0.551、P=0.015;r=0.345、P=0.032)。结论 WNT-1、FZD-1基因蛋白表达下调可能与卵巢癌紫杉醇化疗耐药有一定相关性。Objective: To study the expression and their clinical significance of WNT-1,FZD-1 protein in ovarian cancer tissues and cells after taxol-chemotherapy.Methods: By using immunohistochemical method,the expression of WNT-1,FZD-1 protein in Ovarian cancer tissues and cells were deteced.Among the tissues: 19 cases received taxol-based chemotherapy(group 1) and 39 cases never received chemotherapy before operation(group 2).Cells include sensitive cells and paclitaxel resistant cells.Results: 1.The Integrated option density(IOD) of WNT-1,FZD-1 were significantly 1ower in group1 than those in group 2(30931.34 ±16420.04 vs 45917.65 ±12015.39;38625.20 ±19689.97 vs 55585.82 ±26330.13 respectively,all P 0.05).Observed under light microscope,the protein expression intensity of WNT-1 and FZD-1 in the resistant cells were significantly lower than that in the sensitive cells.2.In group 1 tissues,a significantly lower expression of WNT-1 protein were seen in endometrial adenocarcinoma than that in papillary serous cystadenocarcinoma(19215.88 ± 6506.49 vs 43193.5306 ± 16803.05,P = 0.014);in group 2 tissues,no significant difference in the expression of the two proteins were seen among age,histological types,pathological grade,and clinical-pathological stage.3.The expression of FZD-1 protein was found to have a significant positive correlation with WNT-1 protein(r = 0.479,P = 0.000).In both group 1 and group 2,there was a positive correlation between the expression of WNT-1 and FZD-1 protein(r = 0.551,P = 0.015;r = 0.345,P = 0.032).Conclusions: The down-regulation of WNT-1,FZD-1 genes are related to the mechanism of paclitaxel resistance in ovarian cancer.
