摘  要:目的 观察蛋白激酶 A信号途径对 8-甲氧补骨脂素 (8-MOP)诱导的鼠黑素瘤细胞黑素生成及酪氨酸酶基因表达水平的影响。方法 用半定量逆转录聚合酶链反应技术检测腺苷酸环化酶特异性激活剂毛喉素对 8-MOP诱导的鼠黑素瘤细胞酪氨酸酶活性、黑素含量以及酪氨酸酶基因表达水平的影响。结果 毛喉素与 8-MOP联合处理组所测的酪氨酸酶活性和黑素含量与空白对照组相比提高近 7或 10倍。毛喉素与 8-MOP单独处理组较空白对照组酪氨酸酶 mRNA表达水平上调百分率分别为 26.06%和 6.58%,而联合处理组上调百分率为 56.82%。结论 毛喉素在体外对 8-MOP诱导的黑素生成呈明显增强作用,这一作用与毛喉素在转录水平上调酪氨酸酶 mRNA表达促进酶蛋白合成有关。毛喉素作为治疗白癜风的一种信号转导药物,其临床应用价值有待今后进一步研究。Objective To explore the regulatory mechanism of tyrosinase activity by cAMP/PKA dependent signaling pathway. Methods The regulatory effects on tyrosinase mRNA level were assessed by forskolin, a specific activator of adenylate cyclase in cultured B16F10 murine melanoma cells by using RT-PCR technique. Results It was shown that forskolin could significantly enhance tyrosinase activity, melanin content, and tyrosinase mRNA level. Tyrosinase activity and melanin content were as much as seven to ten-fold higher in cells exposed simultaneously to forskolin and 8-MOP than those in untreated cells. The levels of tyrosinase mRNA were up-regulated by 26.06% with 8-MOP, 6.58% with forskolin, and 56.82% with 8-MOP plus forskolin, respectively. Conclusion Forskolin is shown to enhance 8-MOP-induced tyrosinase activity, probably by way of up-regulation of tyrosinase mRNA expression and tyrosinase synthesis. Further study is needed to understand the therapeutic significance of forskolin for vitiligo.
