人体恶性肿瘤裸鼠移植药敏试验与PCNA表达的相关性研究

摘　　要：目的　探讨人体恶性肿瘤裸鼠移植瘤药敏试验与PCNA表达的内在联系。方法　新鲜实体肿瘤裸鼠肾包膜下移植 3 0例 ,每例 2 4只BALB c裸鼠 ,随机分为 8组 (有 10例只分为 5或 6组 ) ,每组 2只腹腔注射 1种抗癌药 ,1只注射生理盐水对照 ;测定药敏结果后移植瘤行组织学和PCNA检测。结果　移植瘤 12 2 8个 ,其中对照组 40 0个 ,呈正生长60 3 % ,无生长 2 0 8% ,负生长 18 9% ;检测抗癌药 13种 ,敏感 5 1次 ,较敏感 5 2次 ,不敏感 90次 ;送检实验后移植瘤 2 88只 ,光镜下瘤细胞可见率 75 3 % ;PCNA表达阳性率在原始肿瘤标本与对照组中为 69 2 %与 70 8% (P >0 0 5 ) ,在原始肿瘤标本与药敏组中为 69 2 %与 3 9 8% (P <0 0 5 )。结论　人体恶性肿瘤裸鼠移植瘤能保持原肿瘤的生物特性 ;移植瘤药敏组间以及与对照组比较 ,PCNA的免疫标记差异有显著性。提示瘤体对药物敏感度与PCNA呈负相关 ;PCNA指数也与预后有关。Objective To study the relationship between drug sensitive test in nude mouse malignant tumor transplanted from human and PCNA expression.Methods 30 cases fresh tumor from human were transplanted under the capsule of kidney, of to 24 BALB/c nude mice, and mice were divided into 8 groups at random. Two mice in each group were treated by abdominal injection of either anticarcinogen or normal saline. Histology and PCNA were examined after drug sensitive test.Results The number of transplantation tumor was 1 288, 400 in control group, with 60.3% of positive growth, 20.8% of no growth, 18.9% of negative growth. 13 kinds of anticarcinogens were detected, with 51 times sensitive, 52 times fairly sensitive, and 90 times insensitive. 288 transplantation tumor were detected under microscope with anoncocyte visible rate of 75.3%. The positive rate of PCNA expression was 69.2% and 70.8% respectively( P >0.05) in primary tumor samples and control group. Conclusion Malignant transplantation tumor in nude mice human from remains its primary biological characteristics. The immunolabelling of PCNA was significantly different between drug sensitivity group and control sensitivity of tumor to drug is negatively correlated with PCNA, the expression of which is related to porognosis.
