摘  要:目的:采用微小核糖核酸(miRNA)芯片技术筛选出在难治性癫癎患者脑组织中表达有显著差异者,探讨其在难治性癫癎发病中的作用及意义。方法:应用miRNA芯片技术获得6例难治性癫癎患者及4例对照组脑组织中表达有差异的miRNA,筛选出倍数变化(F/C)〉1.5且经t检验P〈0.05并对表达有差异的miRNA进行聚类分析。结果:筛选出表达有显著差异的miRNA基因13条,其中上调基因11条,分别是miR-10a、miR-374c、miR-376a、miR-425、miR-9、miR-224、hsa—miR-26a、let-7c、let-7a、miR-23c及miR-378c,下调基因2条,分别是miR-576—5p及miR-199b-5p。结论:建立了难治性癫癎患者脑组织和非癫癎患者脑组织表达中有差异的miRNA谱,并获得表达显著上调基因11条,显著下调2条,为进一步研究难治性癫癎提供了相关依据。Objective:To obtain differential expression of miRNAs in the brain tissues of patients with intractable epilepsy through miRNA analysis and discuss their effects and significance in the onset of intractable epilepsy. Methods.. Differential expression in brain tissues from 6 patients with intractable epilepsy and 4 controls through miRNA technology, and miRNAs with Fold / Change (F/C)〉1. 5 and P 〈0.05 (t-test) were screened, so as to make clustering analysis of such differential expression. Results: 13 genes of differential miRNAs were found, including 11 genes of up-regulated miRNAs, such as miR- 10a, miR-374c,miR-376a, miR-425, miR-9, miR-224, hsa-miR-26a, let-7c, let-7a, miR-23c and miR- 378e respectively, and 2 genes of down-regulated miRNAs, such as miR-576-Sp and miR-199b-5p respec- tively. Conelusion:A miRNA chart of expression differences was obtained from brain tissues of patients with medically intractable epilepsy and controls. The discovery of 11 up-regulated genes and 2 down-regulated genes proves the research of intractable epilepsy with important and further reference.
