摘　　要：目的建立一种poly(U)加尾RT-PCR检测微RNA(microRNA,miRNA)分子的新方法。方法提取细胞总RNA,用poly(U)聚合酶在miRNA 3'端添加U尾,逆转录为cDNA,实时定量PCR检测miRNA分子。结果经poly(U)聚合酶加尾RT-PCR方法可以成功地检测miRNA分子。结论本研究建立的poly(U)加尾RT-PCR方法可有效地检测并分析miRNA分子,具有特异性强,灵敏度高的特点,为miRNA的检测分析及功能机制研究提供了一种新方法。Objective To establish a new method of poly （U）-tailed RT-PCR to detect microRNA（miRNA）. Methods The total RNA of cells was extracted and polyuridylylated by poly（U） polymerase, then the cDNA was obtained by reverse transcriptase with the universal primer,and the rniRNA was detected by real-time quantitative PCR. Results This method of poly（ U）-tailed RT-PCR could better detect miRNA with specificity and efficiency. Conclusion This method of poly （U） -tailed RT-PCR for detection of miRNAs is simple, economical and efficient.
