摘 要:p53肿瘤抑制蛋白,也被称为基因组监控因子,是一种保护细胞免受一系列生理逆境(例如致癌基因活化、辐射、有丝分裂压力、核糖体压力和化学损害)的转录控制因子。这些生理逆境会导致依赖于p53激活的信号产生,并通过复杂的相互作用网络定位到细胞核,起始转录或抑制许多基因的表达,这些基因与诱导生长停滞、修复、细胞凋亡、衰老或者改变新陈代谢有关。由于p53途径对于调控疾病有如此重要的作用,使得针对该途径的药物干预日益成为人们关注的焦点。本文对调节p53功能的最新研究进展进行综述,并对有关p53基因的癌症治疗前景进行展望。The p53 tumor suppressor protein, also nicknamed the "guardian of the genome", is a transcription factor that protects cells against a range of physiological stresses, such as oncogene activation, radiation, mitotic stress, ribosomal stress and chemical insults. These events lead to signals that are relayed to p53 which gets activated, and gets localized to the nucleus through a complex network of interactions. It turns on a program of transcription or repression of myriad genes that induce growth arrest, repair, apoptosis, senescence or altered metabolism. P53 pathway has such an important role in modulating diseases. This makes pharmacological intervention in the p53 pathway to obtain an increasing focus of attention. We summarized recent developments that attempt to modulate the function of p53 and expected the future of on the p53 gene for anti-cancer treatment.
