摘　　要：目的研究FK506对大鼠面神经损伤后运动神经元凋亡及bcl-2和bax表达的影响。方法将40只Wistar大鼠随机分为实验组和对照组,制作大鼠单侧面神经总干切断模型,实验组每日腹腔注射FK506注射液,对照组给予相同剂量的盐水,在术后各时间点,通过Nissl染色观测面神经核神经元的形态学变化;TUNEL检测细胞凋亡;免疫组化检测bcl-2和bax表达的变化。结果面神经损伤后,FK506组与盐水组相比较,FK506组运动神经元凋亡明显减少,bcl-2表达增加,bax表达降低。结论 FK506有助于增加面神经损伤后运动神经元bcl-2的表达、降低bax表达,抑制细胞凋亡,为药物在神经损伤疾病的临床应用提供了实验依据。Objeelive To investigate effects of FK506 on the apoptosis and the expression of bcl-2 and bax of rat faeial motor neurons af- ter facial nerve injury. Methods A total of 40 Wistar rats were ranldomly divided into experimental and control group. Facial nerve injury model was established by transecting facial nerves al its stylomastoid toramen, and then the experimental group and the control group were treated with FKS06（ FK506-treated group ） and nomlal saline（ saline control group ）by intraperitoneal injection, respectively .The morphology of facial neurons were observed under light microscope at different time points after injury;the apoptotic cells of injured facial motor neurons were detected by TUNEL staining;the expression of bel-2 and bax were evaluated by immunohisochemistry method. Results Mter facial nerve transeetion, the apoptotie cells were, significantly decreased in FKSO6-treated group compared with saline control group （ P 〈 0.05 ） .The expression level of bcl-2 in FKS06-treated group were higher than that in saline control group, and the expression level of bax in FK506-treated group were lower than the control group. Conclusion FK506 could increase the expression of bcl-2,and deerease the expression of l）ax, and inhibit the apoptosis after facial nerve transection .The present results provide an experimental basis for the clinical application of drags in nerve iniury diseases.
