摘 要:目的:明确骨形态发生蛋白-7(bone morphogenetic protein-7,BMP-7)对肝癌细胞株Huh7增殖及迁移能力的影响。方法:往体外培养的肝癌细胞株Huh7中加入重组人BMP-7(recombinant human BMP-7,rhBMP-7),采用CCK-8增殖实验检测Huh7细胞增殖能力,Transwell实验观察肝癌细胞株Huh7的迁移能力;Western blot检测下游信号分子Smad1/5/8蛋白磷酸化水平。结果:CCK-8增殖实验结果显示,rhBMP-7有促进Huh7细胞增殖的趋势,但与对照组相比,差异无统计学意义(P>0.05);Transwell实验结果显示,rhBMP-7能明显减弱Huh7细胞的迁移能力(P<0.05);Western blot结果显示,与对照组相比,加入rhBMP-7后,Smad1/5/8磷酸化水平显著增多。结论:在体外实验中,rhBMP-7有促进Huh7细胞增殖趋势;BMP-7能增加Huh7细胞内磷酸化Smad1/5/8蛋白的表达,并且能促进其核转位。Objective:To investigate the effect of BMP-7 on proliferation and migration of human hepatoma cell line Huh7.Methods:Human hepatoma cell line Huh7 was used.When treated with recombinant human BMP-7(rhBMP-7),the proliferation and migration ability of Huh7 was observed by CCK-8 assay and transwell.The phosphorylation level of Smad1/5/8 protein was detected in Huh7 cell by western blot analysis.Results:CCK-8 assay shows that rhBMP-7 can promote the proliferation of Huh7 cells at 24h,but that has no statistical difference(P〈0.05) in contrast with control group.Transwell assay shows that the migration ability is inhibited in Huh7 treated with rhBMP-7(P〈0.05) by compared with control group.Western blot assay presents that the phosphorylation level of Smad1/5/8 protein was higher in rhBMP-7-treated cells than that in control cells.Conclusions:In vitro experiment,rhBMP-7 has a tendency to promote the proliferation ability of Huh7 cells;the migration ability is inhibited in Huh7 treated with rhBMP-7;BMP-7 can increase the phosphorylation level of Smad1/5/8 protein in Huh7 cells and can promote its nuclear transfer.
