摘  要:目的:探讨脾阳虚状态下模型大鼠体内自由基和体内氧化导致DNA损伤的程度,了解脾阳虚在中医脾病发展过程中的地位,为中医脾阳虚证提供现代科学依据,为中医脾虚衰老学说提供实验依据。方法:首先采用"饮食不节+疲劳过度"复合方法造模14天后,继而给大鼠以苦寒伤阳药灌胃10天复制脾阳虚大鼠模型,取大鼠最后24h尿样测定8-OHdG的含量,取脑皮质和血清测定SOD、GSH-Px、T-AOC、MDA的含量。结果:脾阳虚模型组大鼠血清和脑皮质中SOD活性,GSH-Px活性,T-AOC和MDA含量与正常组相比均有显著差异(P<0.05)。模型组尿样中8-OHdG含量也有明显上调趋势(P<0.05)。结论:脾阳虚模型大鼠体内存在着明显的自由基攻击和氧化损伤,但脾阳虚状态处于脾病全程的何种阶段尚需结合前期脾气虚和脾阳虚实验数据,做进一步探究了解。Objective: This research was to explore the changing rule in the spleen yin deficiency rats'DNA attacked by free radicals and oxidative damage, understanding the spleen yin deficiency in the developmental process of the spleen deficiency, and to provide science evidence for further research and experimental basis for the TCM theories of aging. Methods:Fatty diet + over fatigue method was used to establish the model for 14 days, then the rats were given Shangy- ang drugs for 10 days and the last 24h urine samples from rats were weighed to measure the content of 8 - OHdG, and the SOD, GSH - Px, T - AOC, MDA contents in brain cortex and serum were measured. Results : In serum and cerebral cor- tex, the contents of SOD, GSH - Px, T - AOC and MDA in the model group showed a significant difference ( P 〈 0. 05 ). The 8 - OHdG in the urine samples of the model group was increased significantly ( P 〈 0. 05). Conclusion: There is a clear damage of oxidative and free radical damage in spleen yang deficiency model, but the spleen yang deficiency state in what stage of the spleen disease is needed to understand in - depth studies and combined with previous data of spleen qi and yang deficiency.
