摘  要:目的:通过动态观测急性肠损伤(Acute intestinal injury,AII)肺和大肠等组织中肺表面活性蛋白A(pulmonary surfactant protein A,SP-A)含量的变化,寻找其表达的规律及各组织的相关性。方法:60只SD大鼠随机分为六组:空白对照组、生理盐水组(normal saline group,NS组)、急性肠损伤12h组(AII12h组)、急性肠损伤24h组(AII24h组)、急性肠损伤36h组(AII36h组)、急性肠损伤48h组(AII48h组)。采用结肠内注入醋酸(Ace-tic acid)法制备AII大鼠模型,NS组注入等量NS,空白对照组不作任何处理。分别于注入醋酸后12、24、36、48 h及注入NS后48 h采集标本,空白对照组亦于48 h后采集标本,采用酶联免疫法(enzyme-linked immunosorbentassay,ELISA)检测大鼠BALF及各种组织中SP-A的含量。结果:①各组大鼠BALF中SP-A的含量明显高于肺、大肠、皮肤、肝组织,有极显著性差异(P<0.01);肺组织SP-A含量其次,与其他组织比有显著性差异(P<0.05),小肠组织中无SP-A表达。②NS组、空白对照组BALF、肺、大肠、肝中SP-A的含量较AII36h组高,差异有极显著性(P<0.01)。③AII时间延长,AII各组BALF、肺组织、大肠组织中SP-A的含量下降,至AII36h时含量降至最低;随后有回升趋势。④大肠组织与BALF、肺、皮肤、肝组织中SP-A含量的相关性比较,相关系数r分别为0.292,0.378,0.137,0.518。结论:①大鼠SP-A的合成与分泌具有一定特异性;②BALF、肺、大肠、肝组织中SP-A含量的变化与AII的严重程度有一定的相关性;③大鼠大肠组织中SP-A表达的变化与肝、肺组织中SP-A表达的变化一致性较好。Objective : By observing the content change of SP - A in lung, large and etc in ALI rats, so as to find the regularity expression and the correlation of substance. Methods : 60 SD rats were randomly divided into six groups : blank group, normal saline group, AII12 h group, AII2g h group AII36 h group AII48 h group. The AII rats model was made by instillation with acetic acid directly into the colon,NS group with ns. The rats were killed at 12, 24, 36, 48 hours after modeling to collect samples, ELISA was used for detection of SP - A in all kind of tissues. Results : 1 ) The content of SP - A in BALF was higher than that in lung,large intestine, skin and liver, with very significant difference (P 〈 O. 01 ), and it from lung was followed by BALF, compared to other tissue, with very significant difference (P 〈 0. 01 ). There was no ex- pression in small intestine. 2)The expression of SP -A in NS group and blank group were superior to MI 36 h group, with very significant difference(P 〈0.01 ). 3)With the extension of acetic acid effect, the content of SP -A in BALF, lung and large intestine tissues of AII groups was declined to minimum in AII 36 h group and followed by a rising trend. 4) Through correlation analysis of expression of SP - A, it found that correlation coefficient (r) of large intesine and BALF was 0. 292, r of large intesine and lung was 0. 378 r of large intesine and skin was 0. 137, r of large intesine and liver was 0.518. Conclusion : l )The synthesis and secretion of SP - A from AII rats induce by acetic acid has certain spe- cific;2) the content change of SP- A in lung,large intestine BALF and liver corresponds to the extent of acute intestine injury ;3)the expression of SP -A in large intestine is consistent with lung and liver tissue.
