摘 要:目的:从血液流变学和红细胞膜生物学变化探讨气虚血瘀证在红细胞膜上的生物学差异。方法:20只Wistar大鼠,随机分为空白组及气虚血瘀组,每组10只。气虚血瘀组连续造模15天后,测定血液流变学指标以及红细胞膜组分相关指标。结果:与空白组比较,气虚血瘀组大鼠体质量显著降低;200s-1切变率下,气虚血瘀组全血黏度显著增高(P<0.05);血浆黏度、血沉K值及血浆胆固醇无显著变化(P>0.05),甘油三酯显著降低(P<0.01);红细胞变形指数在600s-1和800s-1切变率下均显著降低(P<0.05),1000s-1切变率下极显著降低(P<0.01);APTT、PT降低极显著(P<0.01);FIB和TT无显著变化;Na+-K+-ATP酶活力及SOD活力均显著降低(P<0.05);MDA含量及膜胆固醇含量有升高趋势但无显著差异(P>0.05)。结论:气虚血瘀模型可能由于红细胞膜上Na+-K+-ATP酶及SOD的减少导致全血黏度及红细胞变形性发生改变,也有可能和APTT、PT等凝血机制有关。Objective: To investigate biological difference of hemorheology of qi deficiency and blood stasis from molecular level of red cell membrane. Method: Twenty wistar rats were randomly assigned to blank con- trol group and qi deficiency and blood stasis group with ten rats in each group. The related indexes of hemo- rheology and the components of erythrocyte membrane were determined after molding 15 days. Result: Com- pared with the blank control group, the weight of qi deficiency and blood stasis group was decreased signifi- cantly. The whole blood viscosity was increased significantly under 200s-1 (P 〈0.05). The red cell deform- ability index was decreased significantly under 600s-1 and 800s-1 (P 〈 0.05 ) while under 1000s-1 (P 〈 0. 01 ). APTr and PT were curtailed significantly (P 〈 0.01 ) while there was no significant difference in TT and FIB. SOD and Na + - K + - ATP enzyme activity of erythrocyte membrane were decreased ( P 〈 0.05 ). The contents of MDA and cholesterol in erythrocyte membrane were increased but there was no significant difference in both of them ( P 〉 0.05 ). The content of hydrosulphonyl was no change ( P 〉 0.05 ). Conclu- sion : The qi deficiency and blood stasis model can decrease SOD and Na + - K + - ATP enzyme activity of e- rythrocyte membrane which may be one of the mechanisms of increasing deformability of erythrocyte and impro- ving the blood viscosity. Maybe there is a relationship with clotting mechanism which includes APTF and PT.
