参黄合剂对内痔组织血管内皮细胞生长因子表达的影响

摘　　要：为观察参黄合剂对内痔组织血管内皮细胞生长因子（VEGF）表达的影响，探讨其治疗内痔的机理。将混合痔60例患者随机分为空白组（生理盐水组）、对照组（高锰酸钾组）和研究组（参黄合剂组），每组20例。空白组患者采用生理盐水坐浴；对照组患者采用1：5000PP液熏洗坐浴；研究组患者采用参黄合剂熏洗坐浴。采用RT—PCR、ELISA法检测应用参黄合剂不同时间段后内痔组织VEGFmRNA、VEGF表达的改变。结果，RT-PCR显示，研究组的内痔组织表达VEGF165mRNA较空白组和对照组少；VEGF蛋白浓度较对照组低，且呈时间依赖性。结果表明，参黄合剂在分子基因、蛋白水平方面均可抑制内痔组织VEGF表达。This study was to observe the influence of Shen-huang mixture on the expression of vascular en- dothelial growth factor（VEGF） of internal hemorrhoid tissue, and explore the mechanism of treatment. The 60 cases with mixed hemorrhoids were randomly divided into normal saline group（as blank group）,PP group（as control group） and Chinese medicine group（as research group） ,20 cases for each group. Patients in blank group was subjected to normal saline sitz bath, while 1 ： 5000 PP solution sitz bath to control group and Shen-huang mixture fuming washing sitz bath to research group. RT-PCR and ELISA meth- ods were adopted to detect the expression change of VEGF mRNA and VEGF at different period after u- sing Shen-huang mixture. As result,in internal hemorrhoid tissue expression, VEGF 165mRNA and protein concentration of VEGF in research group were less and lower than that in control group after treatment with Shen-huang mixture,which showed the time dependence. The results show that Shen-huang mixture could restrain the expression of VEGF of internal hemorrhoid tissue in the aspect of molecular gene and protein level.
