LRP16在子宫内膜样腺癌中的表达分析

摘　　要：目的:探讨正常子宫内膜和子宫内膜样腺癌组织中LRP16的表达及其与人子宫内膜腺癌的相关性。方法:应用RT-PCR技术研究10例正常子宫内膜组织及26例不同分化程度的子宫内膜样腺癌组织中LRP16 mRNA的表达;采用免疫组织化学方法检测上述子宫内膜癌组织中LRP16蛋白产物表达部位及表达水平。结果:子宫内膜样腺癌组织中LRP16mRNA的阳性表达率及表达水平(分别为83.33%、0.82±0.21)明显高于正常子宫内膜组织(分别为30%、0.47±0.18),P<0.05。LRP16在26例子宫内膜癌组织细胞核或胞浆中呈阳性表达,核阳性表达率为73.08%,浆阳性表达率96.15%。在不同分化程度的子宫内膜癌组织中:LRP16核表达与癌细胞的分化程度呈负相关(P<0.05),而LRP16浆表达与癌细胞的分化程度无相关性(P>0.05)。结论:子宫内膜癌组织中LRP16 mRNA的表达明显高于正常子宫内膜组织;且LRP16蛋白产物核表达水平与癌细胞的分化程度呈负相关;过高表达的LRP16与子宫内膜癌的发生和发展可能存在重要联系。Objective： To explore the expressions of LRP16 in normal endometrium and endometrial endometrioid adenocarcinoma and its correlation with human endometrial endometrioid adenocarcinoma.Methods： RT-PCR technique was used to research the expressions of LRP16 mRNA in 10 cases of normal endometrium and 26 cases of endometrial endometrioid adenocarcinoma of different differentiation degrees;immunohistochemical method was used to detect the expression locations and levels of LRP16 protein products in endometrial endometrioid adenocarcinoma.Results： The positive expression rate and level of LRP16 mRNA in endometrial endometrioid adenocarcinoma were 83.33% and（0.82 ± 0.21）,respectively,which were statistically significantly higher than those in normal endometrium 〔30% and（0.47 ±0.18）,respectively〕（P 0.05）.LRP16 positively expressed in cell nucleus or cytoplasm of 26 cases with endometrial endometrioid adenocarcinoma,the positive expression rate in cell nucleus was 73.08%,and the positive expression rate in cytoplasm was 96.15%.Among endometrial endometrioid adenocarcinoma of different differentiation degrees,there was a negative correlation between LRP16 expression in cell nucleus and differentiation degrees of endometrial endometrioid adenocarcinoma（P 0.05）,but there was no correlation between LRP16 expression in cytoplasm and differentiation degrees of endometrial endometrioid adenocarcinoma（P 0.05）.Conclusion： The expression of LRP16 mRNA in endometrial endometrioid adenocarcinoma is significantly higher than that in normal endometrium,but the expression level of LRP16 protein product in cell nucleus was negatively correlated with differentiation degrees of endometrial endometrioid adenocarcinoma;the over-expression of LRP16 may be correlated with occurrence and development of endometrial endometrioid adenocarcinoma.
