摘 要:目的:探讨欧洲花楸悬浮细胞生长规律和动力学参数。方法:测定欧洲花楸悬浮细胞生物量及细胞悬浮液pH值变化;利用细胞鲜重和干重值对欧洲花楸悬浮细胞生长曲线进行拟合,用数学建模模拟欧洲花楸悬浮细胞增殖情况,并对拟合函数求一阶、二阶导数,求算欧洲花楸悬浮细胞生长速率及生长加速度。结果:欧洲花楸悬浮细胞液的pH值随培养时间增加而越来越大,培养时间到16 d,pH>6,不再适宜细胞生长;Logistic模型能够描述欧洲花楸悬浮细胞生长曲线变化。培养第5d时细胞增长加速度达到最大,在8 d时生长速率达到最大,结论:欧洲花楸悬浮细胞生长函数符合logistic曲线,函数为f(x)=0.369 9/(1+10.77e-0.329 2x),细胞在14 d时进入增长平台期,在细胞生长至第5 d时可进行诱导子处理。Objective: We want to make a model of the Cell Suspension Culture of Sorbus aucuparia L. and obtain the Kinetics parameters. Methods:The growth cycle of 20 d and the pH value of ceils suspension liquid of Sorbus aucuparia was measured under normal culture conditions. The growth curve of suspension cultured cells of Sorbus aucuparia was fit by using the fresh weight and dry weight of cells. The growth cycle of suspension cultured cells of Sorbus aucuparia was determined by Logistic function model. Results: The pH of cell suspension increased overtimeobviously during 0 - 14 d. When the pH of suspension cultured cells is over 6, this is not suitable for the growth of cells. The Logistic function model can fit the change of the growth curve of suspension cultured cells of Sorbus aucuparia. The best growth rate and growth acceleration of suspension cultured cells of Sorbus aucuparia is 5 d and 8 d respectively. Conclusion:The function of growth characteristics of suspension cultured cells of Sorbus aucuparia is f(x) =0. 369 9/( 1 + 10. 77e^-0.3292x). Sorbus aucuparia L. cells growth plateau occurson the 14 th day,and reach the fastest speed on the 8th day. Elicitors could be used on the fifth day.
