决奈达隆成盐工艺研究

摘　　要：盐酸决奈达隆为抗心律失常的新药。以决奈达隆为原料,乙酸乙酯为溶剂,滴加盐酸乙醇得到盐酸决奈达隆。考察了成盐溶剂、成盐用盐酸溶液和盐酸乙醇用量对反应的影响,产品经重结晶后纯度达99.99%,收率为48.4%。改进后的工艺避免了使用无水乙酸乙酯和易燃的醚类试剂,降低了生产成本,提高了工业生产的安全性。Dronedarone hydrochloride is a kind of new antiarrhythmic drug.It was synthesized by adding hydrogen chloride ethanol to dronedarone dissolved in ethyl acetate.The effects of reaction solvent,the solvent of hydrogen chloride,the amount of hydrogen chloride in ethanol were examined.The results showed that the purity of the recrystallized product was 99.9% and the yield was 48.4%,the improved salifying process avoided use of anhydrous ethyl acetate and combustible ethers,which had lower production cost and higher production safety.
