摘  要:【目的】为阐明缓慢生长法离体保存野生毛葡萄试管苗的机制,【方法】以野生毛葡萄‘花溪-9’为试材,利用缓慢生长法成功离体保存了供试材料试管苗,并从叶片解剖结构和生理生化变化2个方面阐述了毛葡萄试管苗离体保存存活率与叶片结构和抗氧化酶活性间的关系。【结果】温度、光照和烯效唑(S3307)对野生毛葡萄离体保存效果均有影响,其影响顺序为S3307>温度>光照,且3因素两两之间互作效应明显。野生毛葡萄在低温(10±2)℃、光照10μmol.m-2.s-1(12 h.d-1)和添加0.1 mg.L-1S3307的1/2MS+0.05 mg.L-1IBA+0.1 mg.L-1IAA+3%蔗糖+0.5%琼脂粉的培养基上保存效果良好,存活率可达86.33%。离体保存存活率较高的试管苗叶绿素含量和膜质过氧化产物MDA含量较低,保护酶(SOD、CAT、POD)活性强,渗透调节物质可溶性蛋白含量高,叶肉细胞的紧密度提高,单位面积内细胞数量增多。【结论】适宜的离体保存方法增加了毛葡萄叶肉细胞的紧密度,提高了试管苗的抗氧化能力,这可能是缓慢生长法离体保存成功的重要机制之一。[Objective]In order to clarify the mechanism of in vitro conservation for Vitis heyneana by slowing growth, [Method]the Vitis heyneana 'Huaxi-9' conserved successfully by slowing growth was as material to explore the relations between conservation viability and leaf anatomical structure and antioxi- dant enzymes. [Result] Temperature, light and uniconazole ($3307)all affected in vitro conservation, the effect of $3307 was the greatest, followed by temperature, and that of light was the smallest. There were significant interaction effect between every two variables. 86.33% of in vitro shoots conserved on 1/2MS supplemented with 0.05 mg" L-1 IBA, 0.1 rag. L-1 IAA, 3% sucrose and 0.5% agar, added with 0.1 mg" L-1 $3307 for 10 months at (10_+1) ~C and 10 p, mol "m-Z's-1 (12 h photoperiod) maintained viability. Lower chlorophyll and MDA content and higher protective enzyme (SOD, CAT and POD) activity and soluble protein content were observed on the explant leaves which has high conservation viability. Compared to control, the optimum conservation condition increased cell density and cell number of leaf tissue. [Con- clusion]Increases of cell density and antioxidant activity under appropriate conservation conditions may be the most important mechanism for success of Vitis heyneana conservation in vitro by slowing growth.
