活血化瘀中药对C6脑肿瘤干细胞侵袭能力及TIMP1 mRNA表达的影响

摘　　要：目的:观察活血化瘀中药对C6脑肿瘤干细胞(BTSCs)侵袭能力及组织型金属蛋白酶抑制因子1(TIMP1)mRNA表达的影响。方法:将20只大鼠分为对照组(生理盐水)、B组(桃红四物汤)、C组(血府逐瘀汤)、D组(抵当汤)各5只,给予相应药物灌胃10天后,分离血清分别与C6 BTSCs体外共同培养,进行Boyden体外细胞侵袭试验检测C6 BTSCs侵袭能力变化,荧光定量PCR检测C6 BTCs的TIMP1 mRNA表达变化。结果:B组、C组、D组的C6 BTSCs细胞侵袭率降低、TIMP1mRNA表达升高,与对照组比较,差异均有显著性意义(P<0.05)。结论:活血化瘀中药能抑制C6 BTSCs的侵袭能力,其机理可能在于促进TIMP1 mRNA表达,增强对基质金属蛋白酶的抑制作用。Objective:To observe the effect of blood-activating and stasis-eliminating Chinese herbal medicine on the invasive behavior and the expressions of tissue inhibitor of metalloproteinase-1(TIMP-1)in C6 brain tumor stem cells(BTSCs).Methods:Twenty rats were equally divided into normal control group,Taohong Siwu decoction group,Xuefu Zhuyu decoction group and Didang decoction,and were administered with the normal control and corresponding Chinese medicine orally for 10 days.After treatment,the serum sample of the four groups was extracted for the co-culture with C6 BTSCs.The transwell chambers were used to analyze the invasive capability of C6 BTSCs,and real time PCR was used to detect the expression of TIMP1 mRNA in C6 BTSCs.Results: The three medication groups had lower C6 BTSCs invasive capability and higher TIMP1mRNA expression level than the normal control group,the difference being statistic significance(P < 0.05).Conclusion: Blood-activating and stasis-eliminating Chinese herbal medicine shows inhibitive effect on invasive capability of C6 BTSCs,and its mechanism may be related with the promotion of TIMP1 mRNA expression and the inhibition of matrix metalloproteinase,may interference the invasion of C6 BTSCs directly or indirectly.
