顺铂诱导人肝癌细胞凋亡模型的构建

摘　　要：目的　建立肝癌细胞凋亡模型 ,探讨顺铂抗癌作用机理 ,为进一步研究细胞凋亡分子机制打下基础。方法　用抗癌药顺铂诱导培养的人肝癌细胞株SMMC - 772 1肝癌细胞发生凋亡 ,用四氮甲唑蓝比色试验 (MTT) ,细胞形态学检查 ,DNA凝胶电泳检测。结果　顺铂可显著抑制SMMC - 772 1细胞的生长 ,并有很好的量效和时效关系 ,经药物作用后 ,肝癌细胞在显微镜下呈现典型的凋亡形态学改变 ,DNA琼脂糖凝胶电泳呈现明显的梯度带。结论　用抗癌药顺铂能成功诱导人肝癌细胞株SMMC - 772 1肝癌细胞发生凋亡 ,该细胞凋亡模型的建立 。Objective Establishment of cell apoptotic model of human hepatocellular carcinoma Methods Human cultured hepatocellular carcinoma cell line SMMC-7721 was exposed in vitro to different concentrations of antitumor drug cisplatin for various lengths of time.Cell apoptosis was ascertained by MTT morphology and under electron and light microscopy,DNA gel electrophoresis.Results The growth of cell was suppressed remarkably by cisplatin with a good dose and time response.Typical apoptotic bodies appeared and DNA ladder could be demonstrated on DNA gel electrophoresis.Conclusion The apoptosis of human cultured hepatocellular carcinoma cells,SMMC-7721 cell line,was successfully induced by cisplatin. Establisment of this model will help further explore the molecular mechanism of hepatoma cell apoptosis.
