摘 要:目的研究死亡受体(Fas)和Fas死亡结构域蛋白(FADD)在帕金森病(PD)动物模型不同时相的表达及时程的变化,探讨Fas和FADD在PD发病机制中的作用。方法将40只大鼠随机分为正常对照组(n=8)、假手术组(n=8)和PD模型组(n=24)。PD模型组又均分为PD1、PD2和PD4三个亚组(分别为1周、2周和4周)。检测各组酪氨酸羟化酶(TH)阳性细胞数和Fas、FADD的表达情况。结果模型组TH阳性细胞数均明显减少,Fas和FADD在黑质区域的表达均明显高于正常对照组(P<0.01),但在PD1、PD2和PD4组间无统计学差异(P>0.05)。结论Fas和FADD在黑质纹状体中的持续异常表达可能在PD发病机制中起着重要的作用。Objective To study the expressions of death receptor(Fas)and Fas death domain protein(FADD)in different phases of the rat model with Parkinson′s disease(PD)and to explore the role of FAS and FADD in the pathogenesis of PD.Methods Fourty rats were randomly divided into three groups of blank control(group BC,n=8),sham operated(group S,n=8)and 6-OHDA-induced PD model(group PD,n=24).Group PD was subdivided into three groups of PD1(1week),PD2(2 weeks)and PD4(4weeks).The tyrosine hydroxylase(TH)-positive cell number and the expressions of Fas and FADD were detected.Results The number of TH-positive cells of group PD was significantly decreased,Fas and FADD expressions in the substantia nigra region were significantly increased compared with group BC(P〈0.01),but there was no significant difference among groups of PD1,PD2and PD4(P〈0.05).Conclusion The sustained abnormal expressions of Fas and FADD in nigrostriatal system of rat PD model may play an important role in the pathogenesis of PD.
