摘  要:目的:探讨内外源性胃泌素(gastrin,GAS)对二基肼诱导大鼠大肠癌变中表皮生长因子(epidermal growth factor,EGF)、前列腺素E2(prostaglandin E2,PGE2)表达的影响以及GAS、环氧酶(cyclooxyge-nase,COX)-2、EGF、PGE2、表皮生长因子受体(epidermal growth factor receptor,EGFR)在大肠癌中的作用.方法:140只大鼠随机分为7组:(1)二甲基肼(1,2-dimethyl hydrazine,DMH)+GAS;(2)DMH+奥美拉唑肠溶胶囊(omeprazole entericcapsules,PPI);(3)DMH;(4)DMH+GAS+丙谷胺(proglumide,PGL);(5)DMH+PPI+PGL;(6)DMH+PGL;(7)对照组.放射免疫法检测血清和大肠组织匀浆上清液的GAS、EGF和PGE2浓度,免疫组织化学法检测大肠组织中的COX-2和EGFR的表达,做图象光密度分析.结果:(1)第18周时各组血清和组织中GAS、EGF、PGE2浓度比较:GAS:DMH+GAS(15.59pg/mL±2.90pg/mL,0.38pg/mL±0.11pg/mL)组与DMH+GAS+PGL(15.31pg/mL±5.66pg/mL,0.35pg/mL±0.10pg/mL)组均高于对照组(8.64pg/mL±2.36pg/mL,0.16pg/mL±0.03pg/mL)(P<0.05),DMH+PPI(20.50pg/mL±3.71pg/mL,0.45pg/mL±0.13pg/mL)组与DMH+PPI+PGL(19.90pg/mL±5.10pg/mL,0.37pg/mL±0.11pg/mL)组均高于DMH(13.12pg/mL±3.47pg/mL,0.19pg/mL±0.04pg/mL)组、DMH+PGL(11.45pg/mL±5.13pg/mL,0.20pg/mL±0.05pg/mL)组及对照组.EGF:DMH+GAS(4.26ng/mL±0.92ng/mL,0.011ng/mL±0.005ng/mL)组与DMH+GAS+PGL(4.29ng/mL±0.50ng/mL,0.009n g/m L±0.005n g/m L)组均高于对照组(2.91ng/mL±0.54ng/mL,0.002ng/mL±0.0007ng/mL)(P<0.05),DMH+PPI(5.20ng/mL±1.03ng/mL,0.015ng/mL±0.007ng/mL)组与DMH+PPI+PGL(5.13ng/mL±0.50ng/m L,0.011n g/m L±0.007n g/m L)组均高于DMH(3.76ng/mL±1.47ng/mL,0.004ng/mL±0.002ng/mL)组、DMH+PGL(3.59ng/mL±1.12ng/mL,0.002ng/mL±0.0018ng/mL)组及对照组.PGE2:DMH+GAS(76.03pg/mL±60.75pg/mL,2.74pg/mL±0.76pg/mL)及DMH+PPI(70.29pg/mL±66.58pg/mL,2.42pg/mL±0.89pg/mL)组高于其他组,但差异无统计学意义(P>0.05);(2)腺癌组血清及组织中GAS(32.06pg/mg±15.84pg/mg,0.73pg/mg±0.31pg/mg)、EGF(4.48ng/mg±1.13ng/mg,0.045ng/mg±0.020ng/mg)、PGE2(99.05ng/mg±60.80ng/mg,4.27ng/mg±1.17pg/mgAIM:To examine the effect of endogenous and exogenous gastrin on the expression of epidermal growth factor(EGF) and prostaglandin E 2(PGE 2) in dimethylhydrazine-induced colorectal cancer in rats to explore the role of gastrin(GAS),cyclooxygenase(COX)-2,EGF,PGE 2,and EGF receptor(EGFR) in colorectal cancer.METHODS:One hundred and forty rats were randomly divided into seven groups:DMH+GAS,DMH+PPI,DMH,DMH+GAS+PGL,DMH+PPI+PGL,DMH+PGL,and control group.The concentrations of GAS,EGF and PGE 2 in serum and large intestine tissue homogenate were determined by radioimmunoassay.The expression of COX-2 and EGFR in the large intestine tissue was detected by immunohistochemistry and quantified by optical density analysis.RESULTS:The concentrations of GAS(pg/mL) in serum and large intestine tissue homogenate were significantly higher in the DMH+GAS(15.59 ± 2.90,0.38 ± 0.11) and DMH+GAS+PGL(15.31 ± 5.66,0.35 ± 0.10) groups than in the control group(8.64 ± 2.36,0.16 ± 0.03)(all P 0.05),and in the DMH+PPI(20.50 ± 3.71,0.45 ± 0.13) and DMH+PPI+PGL(19.90 ± 5.10,0.37 ± 0.11) groups than in the DMH(13.12 ± 3.47,0.19 ± 0.04),DMH+PGL(11.45 ± 5.13,0.20 ± 0.05) and blank control groups(all P 0.05).The concentrations of EGF(ng/mL) in serum and large intestine tissue homogenate were significantly higher in the DMH+GAS(4.26 ± 0.92,0.011 ± 0.005) and DMH+GAS+PGL(4.29 ± 0.50,0.009 ± 0.005) groups than in the control group(2.91 ± 0.54,0.002 ± 0.0007)(all P 0.05),and in the DMH+PPI(5.20 ± 1.03,0.015 ± 0.007) and DMH+PPI+PGL(5.13 ± 0.50,0.011 ± 0.007) groups than in the DMH(3.76 ± 1.47,0.004 ± 0.002),DMH+PGL(3.59 ± 1.12,0.002 ± 0.0018) and control groups(all P 0.05).The concentrations of PGE 2(pg/mL) in serum and large intestine tissue homogenate were higher in the DMH+GAS(76.03 ± 60.75,2.74 ± 0.76) and DMH+PPI(70.29 ± 66.58,2.42 ± 0.89) groups than
