结直肠癌组织中miR-142-3p及RAC1基因的表达及其意义

摘　　要：目的研究结直肠癌组织中miR-142-3p、RACl基因及蛋白的表达与临床病理特征的关系，并探讨miR-142-3p及RACl二者表达的相关性。方法用实时荧光定量PCR技术测定50例结直肠癌组织及其对应的正常黏膜组织中miR-142-3p和RAClmRNA的表达；用免疫组化sP法检测RACl蛋白的表达。结果结直肠癌组织中miR-142-3p的表达低于对应的癌旁组织[-（0．714±2．876）比-（2．116±3．179），Z=-2．882，P=0．004]；RAClmRNA表达显著高于癌旁组织[（8．973±1．940）比（10．093-1-2．069），Z=-4．204，P=0．000]。miR·142-3p的表达与结直肠癌的淋巴结转移和临床分期有关（均P〈0．05），而与患者的性别、年龄、肿瘤大小、肿瘤位置、浸润深度、组织分化程度、有无远处转移等无关（均P〉0．05）；RAClmRNA的表达与结直肠癌患者的分化程度、淋巴结转移、远处转移及临床分期有关（均P〈0．05），而与性别、年龄、肿瘤位置、肿瘤大小、浸润深度无关（均P〉0．05）；RACl蛋白的表达与肿瘤的分化程度、淋巴结转移、远处转移及临床分期有关（P〈0．05），而与其他临床病理特征无关。miR-142-3p的表达与RAClmRNA的表达无相关性（P〉0．05），而与RACl蛋白的表达存在负相关（P〈0．05）。结论在结直肠癌组织中RAClmRNA及RACl蛋白高表达和miR-142-3p的低表达可能与其预后有关。Objective To explore the expression and significance of microRNA-142-3p,RAClmRNA and RAC1 protein in the tissues of colorectal cancer. Method Expression of RAN（including microRNA-142-3p and RAC1 mRNA） and RAC1 protein were detected respectively by quantitative real-time RT-PCR, SP immunohistochemical staining in colorectal carcinoma and corresponding normal mucous membrane tissues of 50 cases. Results In colorectal carcinoma, the expression of miR-lg2-3p was lower than that in the adjacent normal tissues [ - （0. 714±2. 876） vs. - （2. 116 ±3. 179） ,Z = -2. 882,P = 0. 004] and the expression of RAClmRNA was significantly higher than that in adjacent normal tissues [ （8. 973 ± 1. 940） vs. （ 10. 093 ± 2. 069） ,Z = - 4. 204, P = 0. 000]. Expression of microRNA-142-3p was associated with lymph node metastasis and clinical stages （ P 〈 0. 05 ）, but no significant differences were observed regarding sex, age, size, site, local invasion, differentiation degree, distant metastasis （ P 〉 0.05）. Expression of RAClmRNA was associated with lymph node metastasis, distant metastasis, differentiation degree and clinical stages （ P 〈 0.05 ）, however, expression of RAC1 protein was only associated with the differentiation degree, lymph node metastasis and clinical stages （P 〈 0.05 ）. There was no statistically significant correlation between the expression of miR-142-3p and RAClmRNA （P 〉 0. 05 ）, except for RAC1 protein. Conclusions In colorectal carcinoma abnormal expression of miR-142-3p, RAClmRNA and protein may have a relevance with prognosis.
