新疆经典型卡波氏肉瘤组织HHV-8与血清巨噬细胞炎症蛋白表达关系的分析

摘　　要：目的:初步探讨在新疆经典型卡波肉瘤(Kaposi’s sarcoma,KS)组织人疱疹病毒8型(human herpesvirus 8,HHV-8)阳性的患者血清中,巨噬细胞炎症蛋白-1α(macrophage inflammatory proteins-1α,MIP-1α)和MIP-1β表达水平差异与KS发病的关系。方法:采用PCR法扩增16例经典型KS肉瘤组织中HHV8ORF26基因片段,证实存在HHV-8病毒。采用ELISA法检测16例KS患者和20名健康对照者血清中MIP-1α和MIP-1β浓度水平。采用2组均数比较t检验来评价其与KS发病的相关性。结果:16例经典型KS患者HHV-8DNA检出率为100%。MIP-1α在KS患者血清中呈现低浓度表达,为(126.72±70.26)ρg/mL;在健康对照组中均呈现高浓度表达,为(1 382.50±448.89)ρg/mL;差异有统计学意义,t=-11.05,P<0.01。MIP-1β在KS血清和健康对照组中呈现高浓度表达,分别为(733.75±276.02)和(886.00±395.65)ρg/mL,差异无统计学意义,t=-1.304,P=0.201。结论:新疆经典型KS患者皮损组织中普遍存在HHV-8DNA病毒,患者血清中MIPs水平的紊乱与新疆经典型KS的发病存在一定的关系。OBJECTIVE： To profile the relationship between macrophage inflammatory protein MIP-la and MIP-I[3 expression levels and clinical significance in herpesvirus type 8-positive tissue from classic Kaposi＇s sarcoma in Xinjiang. METHODS： HHV-80RF26 gene was detected from 16 cases of classic KS patients by using molecular biology test and serum MIP-la and MIP-I~ concentration was detected from 16 cases of KS patients and 20 healthy controls using Enzyme linked immunosorbent assay. The relevances were evaluated using t-test statistical methods. RESULTS： Clinical features has commonality between classic KS of xinjiang and world classic type KS. HHV-SDNA detection rate was 100 ~ from 16 cases of classic KS patients. MIP-I~ showed a low concentration of expression （126.72±70.26）μg/mL in the serum of KS patients and a high concentration of expression （1 382.50±448.89） pg/mL in the healthy control group. Between the two groups there was statistically significant difference （t= --11.05,P〈0.01）. MIP-113 showed high concentrations of expression,with were respectively （733.75±276.02） og/mL and （886.00±395.65） pg/mL in KS serum and in the healthy control group and between the two groups there was no statistical difference （t=- 1. 304, P= 0. 201）. CONCLU- SIONS： HHV-8DNA virus exists widely in lesions of classic KS patients of Xinjiang. There is a certain relationship be- tween the disorder of serum MIPs level and the incidence of classic KS in Xinjiang.
