摘  要:目的探讨香菇多糖对氨磷汀(WR-2721)造成的毒副反应的保护作用。方法香菇多糖和WR-2721作用Hela细胞后,采用MTT法检测其生长情况,采用比色法测定其丙二醛(MDA)含量,采用流式细胞术分析其凋亡情况。结果香菇多糖单独作用Hela细胞时,与空白对照组比较,细胞存活率无显著变化;WR-2721单独作用Hela细胞时细胞存活率显著降低,细胞凋亡率明显增加;香菇多糖与WR-2721联合作用Hela细胞时,细胞存活率显著升高,细胞凋亡率显著降低;香菇多糖、WR-2721单独及两者联合应用时,Hela细胞的MDA含量无显著变化。结论一定浓度的WR-2721对体外培养细胞具有明显的毒性,香菇多糖对WR-2721造成的细胞毒性具有保护作用。Objective To investigate the protective effects of lentinan on the toxic effects induced by WR-2721. Methods After incubation with lentinan and WR-2721, Hela cell growth was detected with MTT assay. The concentration of MDA was determined by colorimetric method and apoptosis was analyzed by flow cytometry. Results When compared with the blank control group, no signifi- cant changes could be noted in cell survival rate of the lentinan group. When WR-2721 activated with Hela cells alone, cell survival rate of the WR-2721 group decreased significantly, while apoptosis increased significantly. When lentinan and WR-2721 inter-activated with Hela ceils, cell survival rate increased significantly and apoptosis rate decreased markedly, while MDA content of Hela cells re- mained basically unchanged. Conclusion WR-2721 at a certain concentration had obvious cytotoxicity on cells in vitro, and lentinan seemed to have protective effects on cytotoxicity induced by WR-2721.
