摘  要:坛紫菜(Pyropia haitanensis)是我国南方沿海的主要海水养殖品种之一,准确的种质鉴定是避免种质混淆和实现其良种化栽培的基础.为筛选出适合于坛紫菜品系间种质鉴定的序列标记,本实验参考动物和植物条形码的构建方法,以11个坛紫菜种质品系为材料,对4种常用于种质鉴定的序列标记(Cox1、Cox2-3、UPA和ITS-5.8S)通过序列同源性分析和构建基于遗传距离的系统进化树两种方法进行比较分析和评价,结果发现:通过序列同源性分析,只有ITS-5.8S序列标记能将11个品系完全分开,而其他3个标记均存在多个品系序列同源性为100%,无法区分的问题;而通过基于遗传距离的系统进化树分析也表明Cox1、Cox2-3和UPA序列标记无法将11个坛紫菜品系完全区分开,而ITS-5.8S序列标记除了8-Ⅰ和9-Ⅳ外,对其他品系均可以很好地区分.由此认为ITS-5.8S是适用于坛紫菜品系间种质鉴定的理想分子标记.Pyropia haitanensis is one of the most important commercial maricuhure species in southern China. For any cultivation, a correct identification of cultivated strains is necessary to ensure a well-bred cultivation and good production quality. To select a reliable marker for discriminating strains of P. haitanensis, 4 frequently used DNA sequence markers (Coxl, Cox2-3 ,UPA and ITS-5.8S) were compared in 11 strains of P. haitanensis with 2 meth- ods. Base on sequence homology analysis, the ITS-5.8S sequence markers are able to discriminate completely 11 strains by comparing their sequences, but the sequences of Coxl, Cox2-3 and UPA are identical and indiscriminate in some of the strains. Furthermore, by comparing the cladograms which are constructed base on K2P genetic dis- tance of each sequence by NJ method, the ITS-5.8S marker can also discriminate 11 strains completely except for 8-I and 9-IV. However, the other 3 markers cannot discriminate the strains because of the existence of 100% se- quence homology. In conclusion, the ITS-5.8S marker is an ideal DNA sequence marker for discriminating strains of P. haitanensis.
