摘 要:目的研究人炎症牙髓组织中核苷酸结合寡聚化结构域2(NOD2)的表达与分布特点,探讨NOD2在牙髓组织天然免疫防御中的可能作用。方法荧光定量聚合酶链反应(qPCR)检测人正常和炎症牙髓组织中NOD2 mRNA的表达情况;采用免疫组织化学染色和免疫印迹方法(Western blot)检测人正常和炎症牙髓组织中NOD2蛋白的定位和表达差异。结果荧光qPCR结果表明,炎症牙髓组织NOD2 mRNA的表达水平显著高于正常牙髓组织;Western blot检测到正常和炎症牙髓组织在96kDa附近均出现蛋白条带,且炎症牙髓组织NOD2表达水平高于正常牙髓组织;免疫组织化学显示,炎症牙髓组织中大量炎性细胞NOD2强阳性表达,成纤维细胞NOD2蛋白也有明显表达,而正常牙髓组织中成纤维细胞未见NOD2蛋白表达。结论 NOD2可能参与牙髓的炎症和免疫反应,推测其可能在牙髓抵御外来病原微生物入侵的宿主免疫防御中发挥作用。Objective To investigate the expression and distribution of nucleotide-binding domain leucine-rich repeat containing receptor nucleotide-binding oligomerization domain 2 (NOD2) in inflamed human dental pulp tissues, and explore the possible role of NOD2 in dental pulp innate immunity. Methods The protein and messenger RNA level of NOD2 in healthy and inflamed human dental pulp tissues were detected by western blotting and fluorescence quantitative-polymerase chain reaction (qPCR). The expression of NOD2 in the pulp tissues were detected by immunohistochemistry. Results The NOD2 mRNA level was significantly up-regulated in inflamed dental pulp tissues compared to healthy ones; NOD2 protein were detected in both healthy and inflamed dental pulp tissues by Western blot, while the expression level was increased in inflamed ones. Immunohistochemistry results demonstrated that NOD2 is strongly positive stained in inflammatory cells and pulp fibroblasts among inflamed dental pulp tissues, while it was not obvious in pulp fibroblasts in healthy pulp tissues. Conclusions These findings suggest that NOD2 participates in the dental pulp inflammatory and immunity response, indicating that NOD2 might play an important role in the immune defense of human dental pulp.
