Immobilization of Candida Rugosa Lipase Enzyme in Organic-Inorganic Hybrid Porous Silica Materials

摘  要:以四甲氧基硅烷(TMOS)和一定比例的甲基三甲氧基硅烷(MTMS)、二甲基二甲氧基硅烷(DMDMS)、三甲基甲氧基硅烷(TMMS)为硅源,采用酸催化的溶胶-凝胶法包埋南极假丝酵母脂肪酶。比较了有机-无机杂化介孔材料和纯硅基介孔材料固定化南极假丝酵母脂肪酶的酶活力和保留活力,同时还比较了有机-无机杂化介孔材料固定化南极假丝酵母脂肪酶和游离南极假丝酵母脂肪酶的pH、温度以及储存稳定性。结果表明,有机-无机杂化介孔材料固定化南极假丝酵母脂肪酶具有比纯硅基介孔材料固定化南极假丝酵母脂肪酶更高的酶活力和保留活力。其中三取代的有机-无机杂化介孔材料固定化南极假丝酵母脂肪酶的酶活力最高,其保留活力可达到84.9%。此外与游离的南极假丝酵母脂肪酶相比,有机-无机杂化介孔材料固定化南极假丝酵母脂肪酶具有更好的pH、温度和储存稳定性,以及更宽的pH值和温度的酶活力范围。Using tetraethoxysilane (TMOS) and organically modified silanes (methyltrimethoxysilane, MTMS; dimethyldimethoxysilane, DMDMS; trimethlmethoxysilane, TMMS)as materials to immobilize Candida rugosa lipase by acid-catalyzedsol-gel method. The enzyme activity and residual activity of Candida rugosa lipase enzyme immobilized organic-inorganic hybrid porous materials and pure silica materials were compared. And the stability of pH, tempe- rature and time of the free Candida rugosa lipase enzyme and the immobilized enzyme were compared. The result showed that the immobilized Candida rugosa lipase enzyme in organie-inorganic hybrid porous material had a higher en- zyme activity and residual activity. The enzyme immobilizedin the tri-substituted alkoxysilanes had a highest enzyme activity and its residual activity was about 84. 9 %. Compared with free Candida rugosa lipase enzyme, the immobilized enzyme had a better stability of pH, temperature and time, and it had a broader pH and temperature range.
