肾癌组织中Wnt5a的表达及其临床意义

摘　　要：目的探讨Wnt5a在肾癌组织中的表达与病理意义以及对肾癌细胞增殖及侵袭潜能的影响,为寻找肾癌有效的治疗途径及预后判定指标提供依据。方法对43例肾癌组织标本和11例正常肾组织标本中的Wnt5amRNA和Wnt5a蛋白表达水平进行检测,分析肾癌组织Wnt5a mRNA表达与临床病理特征的关系,及Wnt5a蛋白表达与预后的关系。结果 Wnt5a mRNA和Wnt5a蛋白在肾癌组织中的阳性表达率均显著高于正常肾组织(P<0.05)。肾鳞癌的Wnt5a mRNA表达水平明显高于肾腺癌(P<0.01);低分化组显著高于高分化组(P<0.01)。Wnt5a蛋白阳性的肾癌患者2年生存率明显低于阴性患者(P<0.01)。结论 Wnt5a表达水平在肾癌细胞中明显增高,且与肾癌的细胞分化程度、组织类型及预后密切相关,提示Wnt5a可能参与肾癌的发生发展过程。Objective To investigate the expression and pathological significance of Wnt5a in renal carcinoma,and its effect on proliferation and invasion potential in renal carcinoma cells,in order to find effective treatment approaches and prognosis indicators of renal carcinoma.Methods The expression of Wnt5a mRNA and Wnt5a protein in 43 cases of renal carcinoma tissues and 11 cases of normal kidney tissues were detected.The correlations between the expres sion of Wnt5a mRNA and clinicopathologic features and between the expression of Wnt5a protein and prognosis in 43 cases of patients with renal carcinoma were analyzed.Results The positive expression rates of Wnt5a mRNA and Wnt5a protein in renal carcinoma tissues were higher than those in normal kidney tissues（P〈0.05）.The expression of Wnt5a mRNA in squamous carcinoma was higher than that in adenometous carcinoma（P〈0.01）.The expression of Wnt5a mRNA in poorly differentiated renal carcinoma cells was higher than that in well differentiated renal carcinoma cells（P〈0.01）.The 2-years survival rate in renal carcinoma patients with positive expression of Wnt5a protein was lower than those with negative expression of Wnt5a protein（P〈0.01）.Conclusion The expression of Wnt5a in renal carcinoma cells increases significantly,and there is significant correlation between it and renal carcinoma cell dif ferentiation,tissue type and prognosis.It suggests that Wnt5a may be involved in the occurrence and development of renal carcinoma.
