摘  要:目的:评价PilC蛋白对人尿道上皮细胞(THUECs细胞)STAT3基因及其下游细胞因子TNF-α和IL-8表达的影响。方法:采用细胞内染色法检测PilC蛋白刺激后THUECs细胞系-磷酸化STAT3(p-STAT3)表达;ELISA法检测细胞培养上清中TNF-α和IL-8水平。结果:PilC蛋白刺激后,细胞内p-STAT3表达升高,且与PilC蛋白浓度成正比,并能增强TNF-α和IL-8的分泌。结论:PiIC蛋白能增强THUECs细胞系内p-STAT3表达和明显增加TNF-α和IL-8水平。Objective:To evaluate the impact of PilC protein on STAT3 gene, and TNF-α and IL-8 ex- pression in urethral epithelial cells (THUECs). Methods: After the THUECs cells were stimulated by PilC protein, the expression of THUECs cells phosphory]ation STAT3 ( P - STAT3 ) was detected by intracellular staining method; TNF-α, IL-8 levels in the supernatant were detected by ELISA.Results: After the cells were stimulated by PilC protein, the intracellular p-STAT3 expression increased, which was positively corre- lated to the concentration of PilC protein. Apart from this, the levels of TNF-α and IL-8 in the supernatant were also elevated. Conclusion: PilC protein can enhance the expression of THUECs cell p-STAT3, and can increase the secretion of TNF-α and IL-8 in cultural supernatant.
