摘 要:目的分析甲状腺激素(TH)在胰岛素抵抗发生、发展过程中对大鼠肝脏脂质代谢的影响,探讨胰岛素抵抗大鼠血清TH水平的变化与胰岛素抵抗大鼠肝脏脂质代谢的相关性。方法 72只雄性SD大鼠随机均分为9组:正常对照组(A组)、高脂高糖(HFS)饲养模型组(B组)、HFS+吡格列酮组(C组,吡格列酮3 mg/kg)、HFS+丙硫氧嘧啶(PTU)低剂量组(D组,PTU 5 mg/kg)、HFS+PTU高剂量组(E组,PTU 10 mg/kg)、HFS+左旋甲状腺素钠(L-T4)低剂量组(F组,L-T450μg/kg)、HFS+L-T4高剂量组(G组,L-T4500μg/kg)、正常饲料+L-T4高剂量组(H组,L-T4500μg/kg)、正常饲料+PTU高剂量组(I组,PTU 10 mg/kg)。各给药组均于实验第1天即灌胃给药,A、B组给予等体积的0.5%羧甲基纤维素钠(CMC-Na)。8周后取血,对各组大鼠空腹血糖(FBG)、血胰岛素(FINS)、血清和肝脏脂质含量及氧化还原指标进行测定。结果 HFS饮食饲养8周可诱导大鼠出现胰岛素抵抗(IR)并伴高血糖、高胰岛素血症、脂质代谢紊乱和氧化还原失衡。与B组大鼠比较,F、G组大鼠脂质分解增加,血清和肝脏中游离脂肪酸(FFA)水平显著升高(P<0.01);肝脏中丙二醛(MDA)含量增加(P<0.05),过氧化氢酶(CAT)减少(P<0.05);高、低剂量的L-T4均明显降低HFS喂养大鼠腹脂指数(AI)(P<0.01),对肝指数(LI)无明显影响。给予PTU后,HFS喂养大鼠总体呈现相反的结果。结论 TH促进HFS诱导的大鼠胰岛素抵抗的形成与发展,促进胰岛素抵抗中肝脏脂质分解代谢。Objective To analyze the effect of different thyroid functional status on hepatic lipid metabolism with the development process of insulin-resistance rats,and to investigate the association and the possible mechanism of thyroid hormone,insulin resistance and hepatic lipid metabolism during the initiation and development of insulin resistance in rats model.Methods 72 Spague-Dawlay(SD) male rats were randomly divided into nine group.Control group(A),high fat and high sucrose(HFS) diet induced insulin resistant group(B),HFS + pioglitazone group(C,pioglitazone: 3 mg/kg),HFS + low/ high dose of Propylthiouracil(PTU) group(D:PTU 5 mg/kg;E:PTU 10 mg/kg),HFS + low/high dose of Levothyroxine sodium(L-T4) group(F:L-T4 50 μg/kg;G: L-T4 500 μg/kg),normal diet + high dose of L-T4 group(H: L-T4 500 μg/kg),normal diet + high dose of PTU group(I: PTU 10 mg/kg).From the first day of the experiment,intragastric administration was given to each treatment group,and at the same time A group and B group were given an equal volume of 0.5% carboxymethylcellulose sodium(CMC-Na).After eight weeks,for all nine groups rats were detected in fasting blood glucose(FBG),fasting insulin(FINS),lipid content and redox indicators in serum and liver.Results Insulin resistant(IR)in rats was induced by HFS diet for eight weeks and it accompanied with hyperglycemia,hyperinsulinemia,lipid metabolism disorders and disbalance of redox state.L-T4 which enhanced thyroid function could result in significant increase of free fatty acids(FFA) in blood and liver(P0.01) due to the enhancement of lipid decomposition in rats of F/G group.L-T4 led to an increase of malondialdehyde(MDA)(P0.05),and was accompanied with a reduction of catalase(CAT)(P0.05) in liver.At the same time the abdominal fat index(AI) was reduced clearly(P0.01) in rats with high/low-dose of L-T4 diet.Chronic PTU treatment on HFS diet induced model rats however showed the opposi
