摘 要:目的结直肠癌患者尿液中5-氟尿嘧啶、奥沙利铂和亚叶酸钙3种常用药物含量的测定。方法对4名结直肠癌患者尿样中5-氟尿嘧啶、奥沙利铂和亚叶酸钙3种药物残留量采用高效液相色谱法进行分离和分析,流速1mL/min,流动相,pH-3磷酸水溶液/甲醇(80/20v/v),检测波长270nm,色谱柱Inertsil ODS(150mm×4.6mm,5μm)。结果5-氟尿嘧啶、奥沙利铂和亚叶酸钙的检出限分别为42.0、294、33.3μg/L,线性范围1.0~1000.0mg/L,相对标准偏差(RSD)分别为0.34%、0.98%、1.00%,回收率93.40%~110.70%。结论该方法样品处理简便,色谱分离完全,结果准确可靠,为结直肠癌患者尿液中的药物代谢组学研究提供了基础。Objective To investigate the detection of the amount of 5-fluorouracil, leucovorin calcium and oxaliplatin in the urine of colorectal cancer patients. Methods The urine of 4 colorectal cancer patients was used by HPLC to separate and analyse the three above-mentioned drugs with flow rate (1 mL/min), mobile phase (phosphoric acid solution/methanol (80/20 v/v)), detection wavelength (270 nm) and column (Inertsil ODS (150 mm × 4.6 mm, 5 tam)). Results The detection limits of 5-fluorouracil, leucovorin calcium and oxaliplatin were 42.0, 29.4, 33.3 μg/L, respectively. The linear range was 1.0-1 000.0 mg/L. The relative standard deviation was 0.34%, 0.98%, 1.00%, and recovery rate was between 93.40 and 110.70%. Conclusion This method is simple, inertsil being fully separated, the result being reliable, which provides a base for future metabolomics research in the urine of patients with colorectal cancer.
