肝肺综合征大鼠肺组织膜联蛋白A2表达的变化

摘　　要：目的评价肝肺综合征（HPS）大鼠肺组织膜联蛋白A2（ANXA2）表达的变化。方法健康sD大鼠40只，雌雄不拘，3～4月龄，体重220～250g。采用随机数字表法，将其分为3组：对照组（c组，n=10）、假手术组（s组，n=10）和肝肺综合征组（HPS组，n=20）。HPS组采用胆总管结扎法建立肝肺综合征模型；c组不做任何处理；S组仅开腹暴露胆总管，但不结扎。术后5周时，处死大鼠，取肺组织，采用RT—PCR法检测ANXA2和平滑肌肌动蛋白α（SM-α-actin）的mRNA表达，采用Westernblot法检测ANXA2和SM—α—actin的蛋白表达。结果与C组比较，S组肺组织ANXA2和SM—α—actin的mRNA及其蛋白表达差异无统计学意义（P〉0．05）；与s组比较，HPS组肺组织ANXA2和SM—α-ctin的mRNA及其蛋白表达上调（P〈0．05）。结论大鼠肝肺综合征时肺组织ANXA2表达上调。Objective To evaluate the changes in the expression of annexin A2 （ANXA2） in lung tissues in rats with hepato-pulmonary syndrome. Methods Healthy 3-4-month-old Sprague-Dawley rats of both sexes, weighing 220-250 g, were randomly divided into 3 groups： control group （group C, n = 10）, sham operation group （group S, n = 10） and hepatopulmonary syndrome group （group HPS, n = 20） . The rats were anesthetized with intraperitoneal 3 % pentobarbital sodium 0.1 ml/100 g. Hepatopulmonary syndrome was produced by chronic ligation of the common bile duct. After 5 weeks, the rats were sacrificed and lungs were removed for determination of ANXA2 and smooth muscle actin a （SM-α-actin） mRNA and protein expression in lung tissues by RT-PCR and Western blot, respectively. Results There was no significant difference in the expression of ANXA2 and SM-α-actin mRNA and protein between groups C and S （ P 〉 0.05）. The expression of ANXA2 and SM-α-actin mRNA and protein was significantly higher in group HPS than in groups C and S （ P 〈 0.05）. Conclusion The expression of ANXA2 in lung tissues is up-regulated in rats with hepato-pulmonary syndrome.
