摘 要:为了提高女贞子提取物中红景天苷的含量,以红景天苷和酪醇的含量为考察指标,采用高效液相色谱法测定其含量,利用正交试验设计优选女贞子提取物中红景天苷的水解工艺,并考察料液比、水解时间和水解温度对提取物中红景天苷含量的影响.结果发现其中冰乙酸与提取液的料液比以及水解时间对提取物中红景天苷的含量起着主要作用;最佳工艺条件为:女贞子浓缩液与冰乙酸体积比为20︰1,100℃下回流20 h.水解后红景天苷的含量约是未经酸水解的3倍.研究表明冰乙酸水解法可显著提高女贞子提取物中红景天苷的含量;该方法操作简单,后续处理容易,水解原料无毒,可以满足大规模工业提取的需要.To develop an acid hydrolysis method to increase salidroside content from the extract of Ligustrum lucidum Ait. fruits, this study, using salidroside and tyrosol contents as indexes, adopted the thogonal design to acquire the optimum conditions for the extraction of salidroside by varying the feed liquid ratio, hydrolysis time and hydrolysis temperature. The contents of both salidroside and tyrosol were measured by HPLC. We found that the feed liquid ratio and hydrolysis time played the leading roles. The optimum processing conditions were: Ligustrum lucidum concentrate : glacial acetic acid = 20:1 (V/V), reflux for 20 hours at 100℃. The content of salidroside after acid hydrolysis was about three times more than that of salidroside before hydrolysis. The results indicated that the glacial acetic acid hydrolysis method can significantly increase the content of salidroside in the extract ofLigustrum lucidum fruits. The acid hydrolysis method is simple to operate, easy in subsequent processing, with the hydrolyzed raw material non-toxic, making it suitable for large-scale industrial production.
