摘  要:目的 研究p16基因改变与脑胶质瘤恶性程度分级及肿瘤细胞增殖活性之间的关系。方法 检测 41例不同级别脑胶质瘤标本和 7例正常脑组织p16基因改变和PCNA蛋白表达情况。结果 实验组p16基因缺失和突变 2 2例 (5 3.7% ) ,mRNA和蛋白表达缺失分别为 2 3例 (5 6 .10 % )、2 7例 (6 5 .85 % ) ,三者改变非一对一关系。p16基因改变和PCNA指数随脑胶质瘤恶性程度级别增高呈上升趋势 (P <0 .0 5 )。p16基因改变与PCNA指数具有明显相关性 (P <0 .0 1)。结论 p16基因改变所导致的细胞周期调控失常及细胞增殖活性升高 ,是影响脑胶质瘤生物学行为的重要因素。Objective To study the relationship between P16 gene modification and the malignant grades of brain glioma and the proliferation activity of tumor cells. Methods The P16 gene modification and PCNA protein expression were detected in 41 brain glioma specimens with different grades and 7 normal brain tissue specimens. Results In the experimental group, the deletion or mutation of P16 gene were found in 53.7% (22/41) cases. The deletion of mRNA and protein expression were found in 56.1% (23/41) and 65.9% (27/41) cases respectively. The change in DNA, RNA and protein levels were without one-to-one relation. The P16 gene modification and increase of PCNA index were related to the increase of malignant grade of brain glioma significantly (P<0.05). Also the P16 gene modification was related to the change of PCNA index dominantly (P<0.01). Conclusion The abnormity of cell cycle modulation and increase of cellular proliferation activity caused by delection or mutation of P16 gene is an important factor to influence the biological characteristics of brain glioma.
