摘  要:目的繁殖和鉴定脆性X智力低下蛋白基因敲除小鼠。方法引进FMR1基因敲除小鼠,提取每只小鼠尾部基因组DNA,用PCR法进行鉴定,获得的雄性纯合子子代小鼠与杂合子雌鼠进行交配。结果 FMR1基因敲除小鼠的饲养和繁殖获得成功,获得较多的基因敲除纯合子小鼠。结论正确的饲养繁殖以及鉴定方法是获得FMR1基因敲除纯合子小鼠的有效途径。Objective To breed and identify fragile X mental retardation 1 (FMR1) gene knockout mice. Methods The FMR1 gene knockout mice were introduced and the genome DNA of tails of mice was extracted and identified by PCR, then male homozygote mice were attained and selected to mate with female heterozygote mice to acquire homozygote baby mice with gene knockout. Results The feeding and breeding were successful and more homozygote baby mice with gene knockout were attained. Conclusion Appropri- ate methods of feeding, breeding and identifying are the effective ways to acquire homozygote baby mice with FMR1 gene knockout.
