薏苡仁水提液对溃疡性结肠炎大鼠血清IL-6、IL-10的影响

摘　　要：目的:观察薏苡仁水提液对TNBS致溃疡性结肠炎(ulcerative colitis,UC)大鼠的作用及对血清IL-6、IL-10水平的影响。方法:Wistar大鼠随机分为6组:空白组、模型组、薏苡仁水提液高、中、低剂量组、柳氮磺吡啶组。TNBS灌肠11 d以产生大鼠UC模型后,灌服相应药物治疗14 d。观察大鼠一般活动状态及大便性状、潜血便血情况,结肠大体形态学及组织病理学改变,进行疾病活动指数评分(disease activ-ity index,DAI)和CMDI评分,酶联免疫吸附法(ELISA)法检测大鼠血清IL-6、IL-10的水平。结果:薏苡仁水提液能显著改善TNBS致UC大鼠的一般状态,减轻其结肠组织病理学损伤。模型组大鼠结肠黏膜DAI比正常组明显增高(P<0.01)。而各给药组较模型组DAI和CMDI均有不同程度的降低(P<0.01),IL-6与模型组相比均有不同程度的降低(P<0.01),而IL-10与模型组相比均有所升高(P<0.01),但薏苡仁水提液低剂量组与其他各给药组相比P<0.01。结论:薏苡仁水提液对UC大鼠损伤的肠黏膜有明显的修复作用,可能通过抑制肠道免疫反应,减少促炎因子和增加抑炎因子的表达,调节促炎因子与抗炎因子间的平衡而发挥其抗炎作用。Objective：To investigate the effects of Coix seed water extraction liquid on expressions of serum IL-6 and IL-10 in rats with TNBS-induced acute ulcerative colitis.Method：Wistar rats were randomly divided into six groups：black group,model group,SASP group,low,middle and high dosage Coix seed water extraction liquid groups.Acute ulcerative colitis was induced by administrating TNBS solution as enema for 11 days.Rats were given drugs by stomach for 14 days.The general morphological and histopathological changes were observed during the experiments for disease activity index score（DAI）and CMDI score.The serum levels of IL-6 and IL-10 were detected by ELISA.Result：In UC rats,the expression of serum IL-6 was significantly decreased whereas IL-10 was increased when compared with the normal group（P〈0.01）.In all treatment groups,Coix seed water extraction liquid could reduce the expression of IL-6（P〈0.01）,increase the level of IL-10 and alleviate the symptom of UC and reduce the DAI.Conclusion：Coix seed water extract on UC rats＇ damage of intestinal mucosa has obvious repair function,may inhibit intestinal immune reaction,reduce promote inflammation factor and increase inhibitory inflammation factor expression,promote inflammation factor and anti-inflammatory factor balance and play its anti-inflammatory effects.
