摘 要:目的探索腺苷预处理对OGD星形胶质细胞NGF和BDNF的影响。方法体外培养SD大鼠大脑皮质星形胶质细胞,将培养至第三代或第四代的星形胶质细胞传代至96孔板内,细胞贴壁并长出突起后,随机分为3组:正常对照组、氧糖剥夺组、腺苷预处理组。观察氧糖剥夺8h复氧糖24h后星形胶质细胞形态、检测细胞培养液中NGF和BDNF的浓度。结果 OGD组星形胶质细胞NGF、BDNF活性明显高于对照组(P<0.05),ADO组与OGD组相比NGF、BDNF活性明显增高(P<0.01),3组间两两比较细胞培养液中NGF、BDNF浓度差异均有统计学意义(P<0.01,P<0.05)。结论腺苷预处理能够进一步增强缺氧缺糖后NGF和BDNF的表达,NGF和BDNF的表达上调可能是腺苷预处理诱导脑缺血耐受,产生神经保护作用的分子机制之一。Objective To explore the effect of adenosine preconditioning on NGF and BDNF in OGD astrocyte. Methods Astrocytes of pallium in SD rat were cultured in vitro. The third or fourth generation of astrocytes was passaged to 96 pore plate. After adherence and evection, they were divided into three groups randomly: normal control group, OGD group and ADO group. The shape of astrocyte and concentration of NGF and BDNF in cell culture fluid were observed at the time of 8 hours after OGD and 24 hours after reoxygenation. Results The vitality of NGF and BDNF in OGD group were obviously higher than that in normal control group(P(0.05). The vitality of NGF and BDNF in ADO group and OGD group were obvi- ously higher(P〈0.01). There were statistical significances in the concentration of NGF and BDNF in cell medium in three groups(P〈0. 01 or P〈O. 05). Conclusion The expressions of NGF and BDNF in astrocyte are higher after OGD. Adenosine preconditioning could increase the expression of NGF and BDNF after OGD. The up-regulation of NGF and BDNF expressions may be one of the molecular mechanism in cerebral ischemia tolerance induced adenosine preconditioning and neural protection.
